黄芪桂枝五物汤对实验性冻疮大鼠血液流变学的影响

【摘要】 目的:考察黄芪桂枝五物汤(五物汤)不同给药途径对实验性冻疮大鼠血流变的影响。方法:采用40%乙醇低温浸泡法制备大鼠冻疮模型,测定模型大鼠及五物汤口服、浸泡、口服+浸泡三种不同给药途径进行干预治疗后血液流变学指标及血栓戊烷B2(TXB2)、6-酮-前列腺素F1α(6-k-PGF1α)的变化。结果:与正常对照组相比,模型组大鼠全血黏度、血浆黏度、红细胞聚集指数及红细胞变形指数显著升高(P<0.05),血清中TXB2显著升高(P<0.05),6-k-PGF1α显著降低(P<0.01)。与模型组相比,五物汤三种给药方式中只有口服+浸泡组可使升高的TXB2显著降低(P<0.05),黄口组与黄口+浸组可使降低的6-k-PGF1α显著升高(P<0.05或P<0.01),且两组间无统计学差异(P>0.05);黄口组、黄浸组、黄口+浸组均可在不同程度上改善大鼠血液流变学指标。结论:冻疮模型大鼠存在微循环障碍及血栓易形成状态,五物汤不同给药途径均有调节冻疮大鼠TXB2-PGF1α失衡、改善微循环血液流变学指标的作用,且不同给药途径中以口服+浸泡给药表现出的药理效应最佳。

【Abstract】 Objective:Study the influence of Huangqiguizhiwuwu Decoction (Hw) on hemorheological behaviors in rats with Chilblains.Methods:Establishing rats’ chilblains model through adopting soaking rats’ feet in low temperature 40% alcohol solution.Determining the contents of TXB2 and 6-k-PGF1α in plasma of the rats with being taken action by Hw through different ways such as oral administration,the immersion,and the oral administration+soak.Results:Compared with the normal control group,blood viscosity,blood plasma viscosity,index of RBC accumulation and index of distortion in rats of model group were significantly increased (P<0.05),TXB2 were significantly increased (P<0.05) and 6-k-PGF1α decreased (P<0.01).Compared with the rats of model group,the increased TXB2 decreased (P<0.05) significantly in plasma of the rats with the Hw taken by the oral administration + soak; the decreased PGF1α increased (P<0.05 or P<0.01) in plasma of the rats with the Hw taken by the oral administration and the oral administration + soak.The Hw groups taken by different ways had improved hemorheological behaviors in varying degrees.Conclusion:the model of Chilblains brings about the unbalance of immunological function,microcycle barrier and a state of forming thrombus easily.The Hw groups taken by different ways can adjust the abnormal changes of immunology,the unbalance of TXA2-PGI2 and improvement microcycle blood rheology targets,and the function of the group of Hw taken by the oral administration + soak is much better than those of the other two Hw groups.
