肺炎克雷伯氏菌灭活疫苗对白鲢的免疫保护效应

【摘要】 以从患烂鳃病的白鲢(silver carp)体中分离的1株致病性肺炎克雷伯氏菌KPCQC01(Klebslella pneumoniae)为材料,采用终浓度为1.0%(v/v)的福尔马林溶液灭活KPCQC01(菌悬液浓度分别为1.4×108CFU/mL、1.4×109CFU/mL、1.4×1010CFU/mL)制备3种不同菌悬液浓度的全细胞灭活疫苗,用该疫苗分别浸泡和注射免疫健康白鲢,然后于第21 d、35 d、49 d、63 d及77 d采集血液,用试管凝集法测定血清抗体滴度,KPCQC01活菌攻毒检测疫苗对鱼体的免疫保护力。结果表明,2种免疫途径均有免疫效果(1.4×108CFU/mL浸泡免疫组除外),且抗体滴度均在第5周达到峰值。其中,菌悬液浓度为1.4×108CFU/mL的菌苗,浸泡免疫后无应答反应,而注射免疫其最高抗体滴度达1:256,免疫保护力为60%;菌苗浓度为1.4×109CFU/mL时,浸泡和注射免疫最高抗体滴度分别达1:64和1:512,免疫保护力分别为40%、75%;而当菌苗浓度提高为1.4×1010CFU/mL时,对应2种免疫途径的最高抗体滴度分别达1:128和1:512;免疫保护力分别为60%、75%。

【Abstract】 To isolate a pathogenic Klebslella pneumoniae(KPCQC01) from a gill disease of risky silver carp as the material,and using a final concentration of 1.0%(v/v)formalin solution inactivated KPCQC01(bacterial suspension concentrations were 1.4×108CFU/mL, 1.4×109CFU/mL,1.4×1010CFU/mL),preparating 3 different concentrations of bacterial suspensions of whole-cell inactivated vaccine,using them to soak and inject the immune health of silver carp.Then in the 21 d,35 d,49 d,63 d and 77 d collect the blood.Using a tube agglutination texts the titer of serum antibody,and KPCQC01 viable attack drug tests the protective immunity in fish of vaccine.The results showed that two kinds of channels are immune(1.4×108CFU/mL immersion immunohistochemistry excluded).And the antibody titers peaked at the 5th weeks.Among them,the bacteria suspension concentration 1.4×108CFU/mL of vaccine don’t have immune responses after immersion.While the injection of immune antibody titer is 1:256 at its highest and the protective immunity is 60%.When the vaccine concentration is 1.4×109CFU/mL pm,the immersion and injection of immune’s highest titer are 1:64 and 1:512 respectively,and the protective immunity are 40%and 75%.When the vaccine concentration increased to 1.4×1010CFU/mL pm,Corresponding to the two kinds of immunity channels,the highest antibody titers are 1:128 and 1:512 respectively,and the protective immunity are 60%, 75%.
