菊黄东方鲀、双斑东方鲀及其种间杂交子代的ISSR分析

【摘要】 通过对菊黄东方鲀、双斑东方鲀及两者的正反杂交子代的ISSR标记分析,研究了2种子代群体之间以及子代群体与共享亲本个体间的遗传关系.使用10个ISSR引物,得到清晰的扩增位点154个,其中多态位点88个,平均多态位点检出率57.14%.分析结果显示:杂交子代个体中具有来自父母本的扩增位点,表明杂交子代确为杂种;杂交子代群体不仅具有较高的遗传多样性,而且在与亲本个体和其他子代群体间的遗传关系上表现出与种内交配子代的明显差异;另外杂交子代与2种亲本的遗传关系上并不对等,表现出明显的倾向性,更加偏向于母本一方。

【Abstract】 Inter-Simple Sequence Repeats(ISSR) was applied to analyse the genetic relationship between Fuguflavidus,Fugu bimaculatus and their two kinds of descendants in order to reveal the genetic differences and the genetic relationships between the parents and their descendants. Ten ISSR primers were used to amplify the genome DNA,154 bands were yielded,88 of which were polymorphic and the average frequency of the polymorphism accoutered for 57.14%.The hybrids inherited bands from both parents,indicating that the hybrids were real heterogenous.By the analysis of the genetic diversity,genetic distance and the loci distributing trend,it was found that the hybrids not only displayed higher genetic diversity,but also displayed distinct difference from t he inbreeding descendants in the genetic relationships with the parents or other descendants.And it was also found that the genetic relationships between hybrids and their parents were not equal,the female parents transferred more genetic loci to their descendants, which indicated that hybrids inherited more genetic information from the female parents or less genetic information from the female parents had changed.
