重力加压状态下成骨的机制及调和气血法的作用研究

【摘要】 目的:观察芪麝丸对长期直立大鼠腰椎骨质增生的作用,探讨药物可能的作用机制。方法:30只SD大鼠随机分为四组,对照组不进行处理,普通饲养笼喂养;模型组和芪麝丸组建立长期直立大鼠模型。用药组在术后8个月开始灌胃,连续灌胃一月,所有动物术后9个月处死,L5椎体藏红染色、天狼猩红染色、免疫组织化学法观察;余下腰椎生长板及椎间盘应用实时荧光定量聚合酶链反应(real time reverse transcript ion polymerass chain react ion,real t ime RT-PCR)技术检测(typeⅠcollegan,ColⅠ的亚型Collα2)、(type X collegan,ColX的亚型Col 10α1)、转化生长因子beta1(transform growth factor beta1,TGF-β1)、血管内皮生长因子(vascular endothelial growth factor,VEGF)、一种核转录因子(core binding factor alpha1,Runx2)基因的表达。结果:藏红染色显示模型组椎体边缘-椎间盘连接处非基质成分增加,发生骨质增生;对照组改变不明显。用药组较模型组比,相同部位非基质成分较少;天狼猩红染色示正常组椎体边缘-椎间盘连接处胶原排列致密,模型组该处Ⅰ型胶原增多,Ⅲ型胶原增多;用药组椎体边缘-椎间盘连接处胶原致密,以ColⅠ为主。免疫组化示模型组骨质增生形成部位Col X,VEGF,TGF-β1表达均较对照组增强,用药组Col X表达均较模型组减少,而TGF-β1表达无明显改变,用药组VEGF表达量较模型组稍有减少。Real time RT-PCR示用药组较模型组下调Col 10α1和Runx2基因表达(P<0.01)。结论:芪麝丸能延缓腰椎边缘-椎间盘连接处骨质增生,与减少X型胶原表达有关。

【Abstract】 Objective To observe the effect of Qi She Wan on long-time upright posture rats’ lumbar vertebral bone formation and investigate its potential mechanism.Methods Tirty SD rats were randomly divided into three groups.The rats had no treatment in control group,and were raised in normal cage.The rats were induced to upright posture in model and Qi She Wan groups. Rats in Qi She Wan groups were given intragastric administration 8 months after surgery.Ten rats were sacrificed at 9 month after surgery and all lumbar vertebras were harvested for detection. Safranine O/fast green staining,picro-sirius red stain and immunohistostaining were performed on L5 vertebrae.Col1α2、Col10α1、TGF-β1、VEGF and Runx2 mRNA expression in other vertebras were detected by Real time RT-PCR.Result Safranine O/fast green staining shows in model group,nonmatrix increased at lumbar vertebrae edge-intervertebral disc junction,as hyperosteogeny happened. However,no obvious change has been observed in the control group.Non-matrix decreased at the same location in treatment group compared with that in model group.picro-sirius red stain shows compact collagens at lumbar vertebrae edge-intervertebral disc junction in control group,however,typeⅠcollagens and typeⅢcollagens increased at the same location in model group.Treatment group showed compact collagens and mainly to ColⅠ.ColⅩ,VEGF,TGF-β1 immunohistochemical positive staining increased at lumbar vertebrae edge-intervertebral disc junction in model group while ColⅩand VEGF positive expression decreased in treatment group compared with those in model one.Real time RT-PCR shows Col10α1 and Runx2 expression was down-regulated in treatment group compared with model one(P<0.01).Conclusion:Qi She Wan may delay hyperosteogeny at lumbar vertebrae edgeintervertebral disc junction,which may be related to reduce type X collagen expression.
