HPGFC法测定沙漠嘎多糖S1的重均相对分子质量

【摘要】 目的:建立以高效凝胶过滤色谱法(HPGFC)测定沙漠嘎多糖重均相对分子质量的方法。方法:通过将沙漠嘎多糖分离、柱层析纯化,获得沙漠嘎多糖S1,采用HPGFC法测定重均相对分子质量。色谱条件:Asahipak GS色谱柱GS-320 7E(7.6mmID×250mmL,9μm)与GS-620 7G(7.6mID×500mmL,9μm)串联使用;流动相:0.003mol/L乙酸钠;流速:1mL/min;柱温:35℃;示差折光检测器(RID)。结果:沙漠嘎多糖重均相对分子质量的回归方程为:1g(Mw)=10.167-0.287t_R(r=-0.976),线性范围为:10 000~500 000Da,测得沙漠嘎多糖S1重均相对分子质量约为8.5×10~3Da。结论:为沙漠嘎多糖重均相对分子质量测定提供了简便、实用的分析方法。

【Abstract】 Objective:To develop a HPGFC method for determine the distribution of weight average molecular weight in the Artemisia haldendron polysaccharide.Methods:Artemisia haldendron polysaccharide S1 was isolated and purified by high performance gel filtration chromatography with refractive index detector.Determination conditions were as follows: Asahipak GS colum:GS-320 7E(7.6mmID×250mmL,9μm) and GS-620 7G(7.6mmID×500mmL,9μm);mobile phase:0.003mol/L sodium acetate;flow rate:1.0mL/min;drift tube temperature:35℃.Results:The regression equation:lg(Mw)= 10.167-0.287t_R(r=-0.976);the linear range of weight average molecular weight of polysaccharide was 10 000~500 000Da.The weight average molecular weight of Artemisia haldendron polysaccharide S1 was 8.5×10~3Da. Conclusion:HPGFC is a practical,reliable method for the determination of the polysaccharide weight average molecular weight.
