RP-HPLC测定5-脂氧合酶抑制剂活性

【摘要】 目的:建立反相高效液相(RP-HPLC)法测定5-脂氧合酶抑制剂活性。方法:采用Brava BDS C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.02%乙酸水,梯度洗脱(0~5 min,比例为22.2:77.8;5~8 min比例由22.2:77.8→15:85;8~18 min,比例为15:85),流速为0.8 mL·min-1;柱温为30℃;分别在235 nm和275 nm检测5-脂氧合酶的主要产物LTB4和5-HETE。结果:LTB4和5-HETE分别在0.02~0.20 mg·L-1(r=0.9999),0.05~0.50 mg·L-1(r=1.0000)范围内呈良好的线性关系;回收率(n=6)分别为100.0%,97.7%及其RSD分别为1.08%和1.52%;日内、日间精密度的RSD分别为0.73%,1.05%和0.26%,1.26%。结论:该法分离效果和重复性均良好,结果准确可靠,可用于LTB4和5-HETE的检测,同时也为5-脂氧合酶抑制剂活性测定提供了可靠的依据。
【Abstract】 Objective:To develop an efficient RP-HPLC method for 5-lipoxygenase(5-LOX) inhibitory activity.Method:The content of LTB4 and 5-HETE,the main products of 5-LOX pathway of arachidonic acid metabolism,was determinded by RP - HPLC method.RP - HPLC was performed on a Brava BDS C18 column(250 mm×4.6 mm,5 um) using a mobile phase consisting of 0.02%acetic acid solution - methanol(gradient elution:0~5 min,22.2:77.8;5~8 min,22.2:77.8→15:85;8~18 min,15:85) at 30℃with the flow rate of 0.8 mL·min-1 and detection wavelength at 235 nm and 275 nm.Result:The calibration curve was linear in the range of 0.02-0.20mg·L-1(r=0.9999) for determination of LTB4,0.05- 0.50 mg·L-1(r= 1.0000) for determination of 5-HETE.The average recoveries(n=6) of LTB4 and 5-HETE were 100.0%(RSD=1.08%) and 97.7%(RSD=1.52%),respectively.The intra-day and inter-day RSDs were 0.73%,1.05%and 0.26%,1.26%.Conclusion:Being sensitive,accurate and reliable with good reproducibility,the method is available for the determination of 5-LOX inhibitory activity.
