丹参对兔视网膜光损伤防治作用的实验研究

【摘要】 目的研究丹参对兔眼视网膜光损伤的防治作用。方法健康青紫兰兔8只随机分为对照组、实验组,每组4只。所有实验动物均在循环光环境适应7d,光照前暗适应24h.实验前,散大瞳孔至8mm左右。光照(14500lux)30min,然后暗适应30min.光照后1h和24h实验动物分别给药。对照组:0.85%生理盐水20ml静脉推注,实验组:丹参冻干粉1.0g/kg+0.85%生理盐水20ml静脉推注。光照后48h取材制成超薄切片,应用电镜观察。结果实验组视网膜组织结构清楚,细胞形态规整。对照组光感受器外节膜盘崩解非常明显,外核层细胞排列紊乱,核固缩。色素上皮层核染色质边集,呈现细胞凋亡的早期改变,线粒体空泡变性。结论丹参对减少兔眼视网膜光损伤方面发挥良好作用。
【Abstract】 Objective A study on the effect of Salvia miltiorrhiza on retinal in rabbits with photic injury. Methods Divide 8 healthy gray rabbits into 2 groups randomly:the control group and the experiment group, with 4 rabbits in each group.The rabbits in each group were subjected to intermittent light exposure for 7 days,with a daily schedule of 12 hours of light exposure and 12 hours of darkness.Then they were given 24 hours of darkness.Before experiment,scatter the pupil till the diameter is 8 mm.The rabbits were given light exposure of 30 minutes(14500lux),then 30 minutes of darkness.After a hour and 24 hours the light injury eyes were administered medicine.In the control group, 0.85%isotonic Na chloride 20ml in vein to inject.In the experiment group,Salvia miltiorrhiza 1.0g/kg + 0. 85%isotonic Na chloride 20ml in vein to inject.After 48 hours eyes were removaled,to make retinal ultrathin sections for observation under elevtron microscopy.Results The rabbits in the experiment group have retinal structures with clear layers and regular cell shape. However,the rabbits in the control group,the outer segments of photoreceptor were arranged in a irregular order, and the stack structure of membrance disks are disorganized,and cells of the outer nuclear layer were in a disorganized pattern,with karyopycnosis.The nuclear chromatin marginatiom emerged in purpurogenous membrane,presented morning changs of apoptotic,and the mitochondria are vesiculated.Conclusion Salvia miltiorrhiza depletes on retinal in rabbits with photic injury.
