山东省分离强毒株猪伪狂犬病毒SA株TK基因结构与功能分析

【摘要】 对山东省分离强毒株猪伪狂犬病毒SA株TK基因进行了克隆和部分序列测定,并分析比较了该序列与国内外主要流行毒株PRV Ea株、Min-A株、LA株、Kroea株、Kaplan株、SH株的同源性。结果表明,核苷酸序列的同源性分别为99.0%、99.1%、98.9%、99.0%、98.3%和74.4%,氨基酸序列的同源性分别为98.7%、98.4%、98.4%、98.7%、98.3%和74.8%。PRV SA株TK具有疱疹病毒胸苷激酶催化结构域的保守氨基酸共有的对应于这两个亚结构域的特征基序。其蛋白的二级结构由45.38%a-螺旋,14.46%β-折叠,6.02%β-转角和34.14%无规则卷曲组成。将PRVTK与鸡、牛、猫科、马科、人类的胸苷激酶用DNA Star绘制进化树分析表明,猪疱疹病毒Ⅰ型TK基因亲缘关系与哺乳动物的疱疹病毒较近,而与禽类的相对较远。
【Abstract】 Thymidine kinase(TK)gene in the unique long region of the wild Chinese Pseudorabies virus(PRV)SA strain was amplified by ploymerase chain reaction(PCR).The PCR product was cloned into pMD18-T vector and sequenced.The nucleotide seqence and amino acid sequence were analysised with DNA Star.The TK gene of PRV SA strain shares highly conserved nucleotides with PRV Ea strain,Min-A strain,LA strain,Kroea strain,Kaplan strain,SH strain.The amino acids of PRV TK contain the conserved ATP-binding motifs(-Y-X-D-G-X-X-G-X-GKS/T-) and-necleosite-binding motifs(-D-R-H-).SOPMA results of the second sructure of PRV SA strain TK was Alpha helix 45.38%,Extended strand 14.46%,Beta turn 6.02%and Random coil 34.14%.The Pro-source relationships between PRV SA TK and chickens,cattle,felidae,equidae,human were studied.Results showed it was closed to mammals,far to fowls.
