全基因组分析禾谷镰刀菌、尖孢镰刀菌和轮枝镰刀菌中的MAPK级联信号途径基因

【摘要】 利用比较生物学方法,从禾谷镰刀菌、尖孢镰刀菌和轮枝镰刀菌基因组数据库中共找出了31个与酿酒酵母菌同源的三类MAPK级联信号途径基因。它们所编码的蛋白质都含有丝氨酸/苏氨酸蛋白激酶结构域,部分还含有SAM结构域。系统进化表明:这三类镰刀菌的MAPK级联信号途径基因分布于10个不同亚群,每个亚群中都含有三类不同的镰刀菌基因。对每个亚群中三类镰刀菌的蛋白质序列和核苷酸序列进行同源性分析,结果显示除亚群Ⅶ同源性低于90%外,其他亚群均高于90%。这说明MAPK级联信号途径基因在这三类镰刀菌中相当保守。依据进化树和同源比对结果对这三类镰刀菌中不同MAPK级联信号网络关系和基因功能进行预测并绘制了信号网络结构图。这些结果将为深入了解禾谷镰刀菌、尖孢镰刀菌和轮枝镰刀菌的MAPK级联信号途径基因功能及其相关信号网络特点奠定基础。

【Abstract】 31 genes were identified from Fusarium graminearum,Fusarium oxysporum and Fusarium Verticillium genome database which homologous to three MAPK cascade signaling pathway genes from Saccharomyces cerevisiae by using the comparative biology method.All of the encoded proteins of these genes contain serine/threonine protein kinase domains,and some also contain SAM domains. Phylogenetic analysis showed that the MAPK cascade genes from Fusarium were distributed into 10 different subgroups and each subgroup contains the genes from three types of Fusarium.The homology of nucleotide and protein sequences of different Fusarium MAPK cascades in each subgroup were analyzed. The results showed that except the subgroupⅦ,the homology in all subgroups were higher than 90%.This results indicated that the MAPK cascade signaling genes were very conservative in three kinds of different Fusarium.Based on the results of phylogenetic tree and homology searching,the function and network structures of the MAPK cascade signaling pathway in different Fusarium were predicted.The results in this paper will facilitate to understand the network structures and functions of MAPK cascade genes in these fungi.
