体细胞突变体筛选法获得象草耐盐植株
Salt-tolerant Plants from Selection of Napier grass Somaclones Mutants

【摘要】 以象草幼穗离体培养诱导产生的胚性愈伤组织为外植体,在愈伤组织继代培养基中添加10~20g/L NaCl,离体筛选得到白色颗粒状愈伤组织,在愈伤组织分化培养基上再生出植株。再生植株耐盐性鉴定结果:6g/L海盐的1/2 Hoagland营养液每隔7d浇灌1次,浇灌4次,处理24d时,对照植株全部致死;处理42d时,24g/L NaCl胁迫筛选获得的MN12-1和MN12-2体细胞再生植株生长良好。取MN12-1、MN12-2植株和对照株的节间嫩芽组培苗进行耐盐盆栽鉴定:4g/L、6g/L海盐的1/2 Hoagland营养液每隔7d浇灌1次,浇灌3次,生长28d时,MN12-1和MN12-2植株的叶、茎、根干重显著高于对照植株,MN12-2植株各部分的干重比对照植株提高100%以上,达到极显著差异。SRAP检测结果表明,MN12-1、MN12-2植株与对照植株存在差异,确认获得了耐盐体细胞突变体。
【Abstract】 The embryogenic calli from immature inflorescences of Pennisetum purpureum were as recipient. White pellet callus were selected by 10~20g/L NaCl added in subculture medium.Regnerated green plants were obtained in the differentiation medium.Evalution the salt tolerance of regnerated green plants showed that All plants of CK were dead after the plants were watered by 6g/L sea-salt in 1/2 Hoagland for four times at 7days intervals and 24 days treatment.The plants of MN12-1,MN12 -2 grew well after 42 days treatment the same as the plants of CK.The regenerated plants,from small shoots of MN12-1,MN12-2 and CK cultured in vitro were evaluated salt tolerance again. Compared with control,dry matter of leaf,stem and root of MN12-1,MN12-2 was significantly increased(P<0.05) after the plants were watered by 4g/1,6g/L sea-salt in 1/2 Hoagland for three times at 7 days intervals and 28 days growth.Leaf,stem and root dry matter of MN12-2 plant in- creased more than 100%in comparison with that of CK plant and reached more significant level(P<0.01).SRAP analysis confirmed that the plants of MN12-1,MN12-2 were different from CK and were salt-tolerant somaclonal variation.
