罗非鱼血浆内氟苯尼考含量的HPLC法

【摘要】 建立了罗非鱼血浆内氟苯尼考的HPLC法。采用内标法和峰高法定量,方法准确性高,重复性良好,并且灵敏度较高;添加氟甲砜霉素标准品到空白样品中,8、1和0.125 g/mL三个浓度的平均回收率为94.59±2.49%;对应的批内精密度为2.78±0.85%;批间精密度分别为3.59±1.90%;检测限达0.03 g/mL。利用本方法对罗非鱼血浆中的药物进行了检测,揭示了28℃淡水条件下,罗非鱼以剂量10mg/kg口服氟苯尼考后10h内血浆中的药物浓度变化规律。

【Abstract】 A liquid chromatographic(HPLC)method for determination of florfenicol in the plasma of Tilapia was developed.For quantitation,the peak-height measurements and the internal standard was used. The study showed that the linearity and repeatability is good.The method was validated with blank samples fortified at 8,1 and 0.125 g/mL,the mean recovery was 94.59±2.49%,the coefficient of variation was below 6.3%,and the detection limit for flofenicol is 0.03 g/mL,otherwise,the law of the variation of the drug plasma concentration was disclosed,by using the method to determine the florfenicol within 10 hours in Tilapia after oral administration of the drug in freshwater at 28℃.All the results indicate its usefulness for conducting pharmacokinetics studies on Tilapia.
