贵州省鼠疫耶尔森氏菌生物学特征研究

【摘要】 目的研究贵州鼠疫耶尔森氏菌的生物学特征,确定疫源地性质。方法对37株鼠疫菌进行生化试验、营养需求试验、毒力基因检测、随机扩增多态性DNA分析和脉冲场场电泳分析。结果37株鼠疫菌均不发酵鼠李糖和甘油,发酵麦芽糖和阿拉伯糖,脱氮阳性。20株代表菌株均为苯丙氨酸依赖(Phe~+)、谷氨酸半依赖(Glu~±);用Pla、Cafl、inv、hms四个基因设计引物对20株代表菌株分别扩增,均得到456bp、249bp、1000bp和700bp的条带;随机引物对20株代表菌株分别扩增,均获得505bp、790bp、1140bp和1680bp的条带;20株代表菌株的脉冲场电泳图谱显示338kp、242.5kp、168kp和77kp的条带。结论贵州鼠疫菌株的生物学特征与我国滇闽居民区型鼠疫菌相同。

【Abstract】 Purpose In this study,the biological characteristics of yersinia pestia in Guizhou were described and the features of plague foci,where the plague broke out,were determined.Method 37 strains of yersinia pestia were studied at different levels,including biochemistry experiment,nutritional requirement,toxicity gene detection,random amplified polymorphic DNA(RAPD) analysis and pulsed field electrophoresis analysis.Result 37 strains of yersinia pestia do not wholly ferment rhamnose and glycerol,but ferment maltose and arabinose. They are all denitrification-positivity.Twenty typical strains are distinguished by Phe~+ and Glu~±. When the 20 typical strains were amplified with the primers,which were designed on the basis of 4 genes:Pla,Cafl,inv and hms,the DNA fragment of 456bp,249bp,1000bp and 700bp were all amplified.When the 20 typical strains were amplified with the random primer,the DNA fragment of 505bp,790bp,1140bp and 1680bp were all amplified.The pulsed field electropherograms of the twenty typical strains show that the DNA fragment of 338kp,242.5kp,168kp and 77kp were amplified.Conclusion The biological characteristics of yersinia pestia in Guizhou are congruent with the ecatype of residential areas of westem Yunan,Fujian and Guangdong in China.
