黑龙江省甜瓜白粉病病原菌鉴定及rDNA-ITS序列分析
Identification and Sequencing of Ribosomal DNA-ITS of Pathogen of Melon Powdery Mildew in Heilongjiang

【摘要】 首次应用通用引物ITS1和ITS4从采自黑龙江省3个地点的甜瓜白粉病菌株中扩增出目的片段为524bp的rDNA-ITS序列(GenBank登录号为EU294368)。与GenBank核酸序列数据库进行BLAST比对,结果表明,供试菌株与叉丝单囊壳属Podosphaera fusca(登录号:EF137855、EF137859、EF137853)的ITS序列完全相似,同源性为100%。采用考马斯亮蓝组织透明染色方法对该病原菌进行形态鉴定,可以清楚观察到白粉病菌体的5个发育阶段,其形态特征为:分生孢子椭圆形、无色、串生、不分枝、平均长度为29.07μm、平均宽度为17.82μm。接种在鉴别寄主瓠瓜上表现为感病。形态学、鉴别寄主和分子鉴定结果证明,黑龙江省甜瓜白粉病病原菌为Sphaerotheca fuliginea。
【Abstract】 DNA was extracted from the strain of pathogen of melon powdery mildew using a modified CTAB method.ITS sequence (524 bp) was initially amplified from the pathogen by using the universal primers ITS1 and ITS4 (registered No.EU294368).Comparing to the nucleotide sequences acquired from GenBank database,the strain is clustered into the homogeneity with Podosphaera fusca (EF137855、EF137859、EF137853),with a homology of 100%.The different phases in life cycle of Sphaerotheca fuliginea were observed through the method of coomassie brilliant blue staining clearly,included germinating conidia,primary germtube,hyphae,colidiophore and colony. Morphologic characteristics of Sphaerotheca fuliginea were conidia ellipse,colorless,catenulate,average length: 29.07μm,average width:17.82μm.The identifying host showed infection.Results of morphology,molecular identification and the identification host showed that pathogen of melon powdery mildew in Heilongjiang was Sphaerotheca fuliginea.
