布地奈德对哮喘大鼠气道嗜酸细胞影响机制的实验研究

【摘要】 目的观察布地奈德(Bud)对哮喘大鼠气道嗜酸性粒细胞趋化因子(Eotaxin)的表达及嗜酸细胞(EOS)在气道局部浸润的影响。方法复制哮喘大鼠模型,分为正常组(C 组)、哮喘组(A 组)和 Bud 组(B 组),用免疫组化检测肺组织 NF-κBp65亚基及 Eotaxin 蛋白活性,并将结果以图像分析系统加以量化;细胞分类计数法检测支气管肺泡灌洗液(BALF)中 EOS 的绝对计数和百分比;病理学方法观察气道炎症程度。结果 A 组肺组织 NF-κBp65(0.256±0.063)及 Eotaxin(0.197±0.055)表达量均显著高于 C 组(分别为0.015±0.006,0.034±0.008)(均为 P<0.01)。 B 组肺组织 NF-κ Bp65(0.062±0.014)及 Eotaxin(0.070±0.053)的表达均显著低于 A 组(均为 P<0.01):B 组 BALF 中 EOS 的绝对计数和百分比均低于 A 组(均为 P<0.01);光镜下观察 B 组气道炎症较 A 组显著减轻。结论哮喘大鼠肺组织 NF-κBp65及 Eotaxin 表达显著增强,Bud 能抑制 EOS 在气道局部的浸润,减轻气道炎症。其机制可能与抑制哮喘大鼠肺组织 NF-κB的活性从而减少 Eotaxin 的转录合成有关。

【Abstract】 Objective To study the role of budesonide(Bud)in regulating the activity of NF-κ Bp65 and Eotaxin protein of asthma rats,and in effecting the infiltration of eosinophil within airway.Method Thirty-six Sprague-Dawley(SD)rats were divided three groups of 12 each.The asthmatic model was established by ovalhumin(OVA)injection and inhalation.Group B were pretreated with Bud atomized inhalation(2mg/d)within each of 18 days.The protein NF-κ Bp65 and Eotaxin in lung tissue were assessed by using immunohistochemisty and quantify the results into light density by the analyse system of medical image:The numbers of EOSand the EOS% in BLAF were counted by different count fluids:The condition of airway inflammation were observered by pathology Results Twenty-four hours after the last antigen challenge,the protein expression of NF-κ Bp65 (0.256±0.063)and Eotaxin(0.197±0.055)in group A were significantly increased compared with group C(0.034±0.008:0.010±0.005,p<0.01,respectively).The protein expression of NF-κ Bp65(0.062±0.014)and Eotaxin(0.070±0.053)in group B were significantly decreased compared with group A(p<0.01,respectively):The numbers of EOS and the EOS% in BLAF of group B were significantly decreased compared with group A(p<0.01, respectively);The conditions of airway inflammation of group B were significantly decreased compared with group A under light microscope.Conclusion In asthmatic rats, the protein of NF-κ Bp65 and Eotaxin were increased significantly,Bud could reduce the infiltration of eosinophil within airway and decrease the condition of airway inflammation;The mechanism is possibly that the activity of the protein of NF-κ Bp65 was inhibit ed,then the transcription of Eotaxin was hold down.
