血脂康对大鼠在体心脏急性缺血—再灌注心肌Fas/FaSL表达的影响

【摘要】 目的:探讨血脂康对大鼠在体心脏急性缺血—再灌注心肌 Fas/FasL 表达的影响。方法: 40只雄性 SD 大鼠随机分为5组:假手术组(Sham group,S,n=8),双蒸水灌胃一周;模型对照组 (Control group,C,n=8),双蒸水2ml 灌胃一周;甲羟戊酸组(Mevalonate group,M,n=8),甲羟戊酸150mg/Kg.d 溶于2ml 双蒸水灌胃一周;血脂康组(Xuezhikang group,X,n=8),血脂康2.8g/Kg ·d溶于2ml 双蒸水配成悬液灌胃一周;血脂康+甲羟维酸组(X+M,n=8)。血脂康2.8/Kg.d 联合甲羟戊酸150mg/Kg·d 溶于2ml 双蒸水灌胃一周。一周后应用0000无创丝线结扎冠脉前降支 30min,随后松开复制在体心脏急性缺血—再灌注模型,假手术组仅穿线而不结扎前降支,其他手术步骤相同。再灌注120min 后观察各组心肌 Fas 和 FasL mRNA 及蛋白表达的情况。结果:与 C 组 Fas/FasL 表达相比较,X 组 Fas/FasL 表达明显减少(P<0.01);而 X+M 组与 X 组相比较 Fas/FasL 表达显著增加(P<0.01);C 组、M 和 X+M 组三者之间相比较无差异(P>0.05)。结论:血脂康能减少心脏急性缺血—再灌注心肌 Fas/FasL 的表达,血脂康抑制心肌中 Fas/FasL 的表达同血脂康中含有的他汀类物质有关。

【Abstract】 Objective:To explore the effects of Xuezhikang on the expression of Fas/FasL in myocar- dium during rats heart isehaemia-reperfusion in vivo.Methods:40 Sprague-Dawley male rats were ran- domly divided into five groups:sham group(S),rats had intragastric administration with 2ml ddH20 once a day;modle control group(C),rats had intragastric administration with 2ml ddH20 once a day; mevalonate group(M),rats were lavged with mevalonate 150mg/kg solving in 2ml ddH20 once a day for one week;Xuezhikang group(X),rats were treated with Xuezhikang 2.8g/Kg solving in 2ml ddH2O once a day for one week;Xuezhikang+mevalonate group(X+M),rats were treated with Xuezhikang 2.8g/kg and mevalonate 150mg/Kg solving in 2ml ddH2O once a day for one week.The myocardial is- chaemia-reperfusion modle was produced by ligating the left coronary artery 30min and relaxation to reperfuse 120min after one week administration with 0000 silk suture in vivo.In the sham group,sham op- erations(with 0000 silk suture put beneath the artery but without ligation)were performed.The expres- sion of Fas/FasL were detect by RT-PCR and western blotting.Results:Compaired with C group,X group expressed Fas/FasL decreasly(P<0.01).The expression of Fas/FasL incresed in X+M group (vs X group,P<0.01).While Fas/Fasl expressions were not different between C group,M group and X +M group(P>0.05).Conclusion:Xuezhikang can reduce the expression of Fas/FasL in myocardium during rat heart ischaemia-reperfusion in vivo and mevalonate can inhibit these effects.
