【摘要】 目的:观察人参环氧炔醇(panaxydol,PND)对10%胎牛血清(FBS)诱导的大鼠主动脉平滑肌细胞(RASMCs)迁移和线粒体转录因子1 mRAN 表达的影响。方法:采用细胞刮片法观察 RASMCs 的迁移,MTT 法观察 RASMCs 的增殖,比色法观察羟脯氨酸含量,RT-PCR 观察线粒体转录因子1(mitochondrial transcription factor 1,mtTF1,mtTFA)mRNA 的表达。结果:PND 以浓度依赖方式抑制10%FBS 诱导的 RASMCs 迁移、增殖和基质合成,同时能降低10%FBS 引起的 mtTF1 mRNA 表达上调。结论:PND 具有抗 RASMCs 迁移、增殖和基质合成的作用,该作用与抑制 mtTF1 mRNA 表达有关。

【Abstract】 Objective:To study the effects of panaxydol (PND) on the migration and expression of mtTF1 mRNA in rat aortic smooth muscle cells (RASMCs) induced by 10% FBS were investigated.Methods:The effects of different concentrations of PND on the migration and prolifera- tion of RASMCs were evaluated by cell scrape and MTT tests.The content of hydroxyproline was observed by colorimetry method.The influ- ences of PND on the expression of mtTF1 mRNA was studied by RT-PCR.Results:1,3,9μmol/L of PND could inhibit RASMCs migra- tion,proliferation and the content of hydroxyproline induced by 10 % FBS.And it could suppress the up-regulation of expression of mtTF1 mRNA in RASMCs.Conclusion:PND could inhibit the RASMCs migration,proliferation and matrix synthesis induced by 10% FBS,and de- crease of mtTF1 mRNA expression might be one of its mechanisms.
