蓝色犁头霉发酵法制备甲壳素和壳聚糖的研究

【摘要】 对以犁头霉发酵法制备甲壳素和壳聚糖的工艺条件进行了研究,优化出较佳的工艺条件和方法,并进行了培养基和发酵条件的单因素与正交试验,结果表明:蓝色犁头霉较佳培养基组成为:蔗糖2.5%,牛肉膏1.2%,K2HPO4 0.075%,生物素0.0003%;其较优发酵条件为:温度25℃,周期48h,pH 6.0,摇瓶装液量75 mL/250 mL。但蓝色犁头霉发酵法制备甲壳素及壳聚糖的产量不高,与用化学法相比并不经济,要实现工业化生产,尚需寻找新菌种或采用基因工程、诱变育种等手段以制造出高产甲壳素和壳聚糖的遗传性状稳定的工程菌。
【Abstract】 The methods and technology of preparing chitin and chitosan by fermentation with Absidia coerulea was studied.Single factor and orthogonal experiments for medium and fermentation condition were carried out in the experiment.The results indicated that the optimal composition of fermentation medium for Absidia coerulea was:sugar 2.5%,beef extract 1.2%, K2HPO4 0.075%,biotin 0.0003%.The optimal condition of fermentation was:temperature 25 ℃,time 48 h,pH 6.0,amount of medium per flask 75 mL/250 mL.However,the yield was lower than chemical extraction by now.To make this method into industrialization,there is need for new engineering fungi which has high productivity and stable genetic properties using breeding technologies such as mutation and gene engineering.
