基于电聚合蛋白A固定抗体的免疫微传感芯片

【摘要】 采用 MEMS 工艺在硅衬底上制备了电极型免疫微传感芯片,重点探索了微传感器敏感表面抗体固定化的方法。尝试采用蛋白 A 与吡咯共同电化学聚合,进而由蛋白 A 结合抗体的方法,实现了抗体的固定。将该免疫传感器对人免疫球蛋白 G(HIgG)进行检测,在5~160 ng/mL 以及 160~640 ng/mL 呈分段线性的特性,实现了微剂量、低浓度的免疫检测,验证了该固定化方法的可行性。此法具有省时、简便、易控、适合微传感器固定的优点。

【Abstract】 An immunosensor of microelectrodes was fabricated on silicon wafer using MEMS technology.An antibody immobilization method suitable for micro scale was developed.Protein A and polypyrrole were electropolymerized,and antibody was immobilized by the oriented bond with protein A.Human immunoglobulin G(HIgG)was detected by this micro immunosensor with low concentration and small reagent usage.Two linear responses to HIgG concentrations from 5 ng/mL to 160 ng/mL,and from 160 ng/mL to 640 ng/mL,respectively,were displayed by this micro immunosensor.The feasibility of the antibody immobilization method was proved. This immobilization method shows many advantages,such as fast,simple,controllable and suit- able for microsensor.
