基于表达序列标签(EST)策略的茶树分子生物学研究

【摘要】 表达序列标签是在人类基因组计划实施过程中,由美国国立卫生研究院生物学家Venter提出的发现基因的新战略。EST技术已被广泛应用于新基因的克隆、组织特异性基因表达谱分析以及基因组序列的功能注释等许多研究领域,它已成为大规模获取功能基因的一种最为快捷和有效的研究手段。茶树是一种重要的木本经济植物,其分子生物学研究起步较晚,大大滞后于禾谷类与许多其他木本植物。最近,基于cDNA文库和EST测序分析技术的茶树分子生物学研究,包括基因表达谱特征分析、cDNA芯片建立与应用、EST-SSR/STR标记开发与应用、次生代谢以及与抗逆相关基因的分离克隆和表达分析等,取得了一些重要的进展,对加快探索和认识茶树的生育、代谢和抗逆等的分子机理等都具有积极的意义。

【Abstract】 Expressed Sequence Tag (EST) was developed with the progress of Human Genome Project by the biologist Venter from the Natural Institute of Health of the United States of America, and it is a new and robust approach to hunt for new genes. ESTs technique provides a quick, efficient and inexpensive route for novel gene cloning, gene expression and regulation analyses, genome mapping and functional annotation for the genome sequence. Tea plant is an important cash woody species. Its molecular biology research initiated much later and progressed rather slower compared with cereal crops and some other woody species. Most recently, remarkable progress on molecular biology of tea plant based on the strategies of cDNA library and EST sequencing and analyzing have been made, including elucidating gene expressing profiling, establishing and utilizing of cDNA microarray, data mining for EST-SSR/STR, cloning and expressing and analyzing of genes involved in secondary metabolism and stress defense, etc. All these contribute to quickly explore and understand the molecular mechanism of the growth, metabolism and stress defense of the tea plant, as well as to promote the global research level of molecular biology of tea plant, a perennial woody plant species.
