山楂叶总黄酮提取纯化工艺研究

【摘要】 目的:优化山楂叶总黄酮提取纯化工艺。方法:通过比色法进行总黄酮含量测定,采用正交试验设计,对乙醇提取工艺及 HP-20、HPD-200、HPD-500型大孔吸附树脂纯化工艺进行筛选和优化。结果:最佳提取工艺:采用70%乙醇提取三次,70℃的水浴加热,每次用10倍量的乙醇提取1h。在此提取工艺条件下,总黄酮得率最高;最佳纯化工艺:用 HPD-500 型大孔吸附树脂进行吸附,70%乙醇洗脱。在此纯化工艺条件下,山楂叶总黄酮纯度达到85%以上,且转移率达到72%以上。结论:优化确定了山楂叶总黄酮提取纯化工艺。

【Abstract】 OBJECTIVE:To determine the optimal parameters for the extraction and purification process of total flavonoid from Hawthorn leaves.METHODS:Total flavonoid of Hawthorn leaves was determined by colorimetry.It was investigated by the orthogonal experiment design of form L9(34) with the extraction process by alcohol and the purification process by screening three types of macroporous resin,which were HP-20,HPD-200 and HPD-500.RESULTS:The optimal condition of extraction was with 70% alcohol as much as ten times of medical material amount and was bathed for 1 h and three times in 70℃ water.Under these conditions,the extraction ratio of total flavonoid from Hawthorn leaves was highest.The optimal condition of purification was as follows:HPD-500 macroporous resin as absorbents,and 70% alcohol as eluant.Under these conditions,the purification ratio of total flavonoid from Hawthorn leaves was more than 85%.The transition ratio was more than 72%.CONCLUSION:The optimal parameters for the extraction and purification process of total flavnoids from Hawthorn leaves were determined.
