【摘要】 以非抗螨、非抗蚜陆地棉品种(系)及3个种外材料为对照。利用 RAPD 标记技术对育成抗螨、抗蚜棉花品种(系)进行遗传差异性分析。从80条中筛选出17条能揭示供试材料遗传多态性的 RAPD 引物,17条引物共扩增出了93条带,其中具有多态性的有85条。采用 Jaccard’s 相似系数, 非加权维平均法(UPGMA)聚类,结果表明,16个棉花材料基本按照抗螨、抗蚜、种外材料分别聚在一起。材料之间的相似系数变幅较大,遗传相似系数在0.254～0.910。其中抗螨材料与抗蚜材料之间存在一定的遗传差异(0.603～0.819)。聚类结果分析及遗传相似系数分析与材料来源的系谱相符,为抗虫育种的种质资源利用与棉花抗螨、抗蚜的分子标记辅助育种提供指导。更多还原

【Abstract】 RAPD analysis was used to assess the genetic variation of some cotton germplasms for re- sistance to red spider (Carmine spider mite) and aphid (Aphis gossypii),with comparisons of other important upland cotton varieties susceptible to red spider and aphid,as well as Gossypium barba- dense,G.gossypioides,G.thurberi lines.17 RAPD primers selected from 80 were used to reveal genetic diversity.Total 93 bands were obtained in all tested lines,and 85 of them exhibited poly- morphism.Jaccard’s similarity and UPGMA cluster analysis showed that the genetic similarity a- mong them ranged from 0.254 to 0.910.There are some genetic differences between cotton varie- ties resistant to red spider and to aphid(0.603～0.819).G.barbadense,G.gossypioides,G.thurb- eri were clustered independently as expected.The results of the cluster analysis and the genetic sim- ilarity were consistently with the pedigree information of the 16 cotton varieties.This research pro- vided a guide for using the pest resistant germplasms effectively,and for further molecular marker- assisted selection for cotton resistance to red spider and aphid.更多还原
