东方百合试管鳞茎生产技术研究
Study on Bulblet Production in Lilium Oriental Hybrids

【摘要】 以东方百合不同品种脱毒组培苗为材料进行试管鳞茎膨大培养研究,结果表明,东方百合试管鳞茎膨大及休眠特性品种间存在差异,提高蔗糖浓度至90g/L 可促进鳞茎休眠。甘露醇对试管鳞茎品质提高有明显的促进作用。磷用量为 MS 标准用量的1.5和2.0倍明显促进根系和愈伤组织生长,但不利于鳞茎的膨大。采用5℃低温处理对打破鳞茎休眠效果明显,处理时间以45~60d 为宜。采用鳞茎芽为外植体培育试管鳞茎, 经10~15℃预处理40d,再在5℃处理50d,试管鳞茎抽茎率可达37.8%。
【Abstract】 The plantlet from different Lilium oriental hybrids were used as explant for bulblet enlargement culture in vitro.The resuhs showed that bulblet enlargement and dormancy development were difference between varieties.The bulblet dormancy de- velopment was increased with high level of sucrose concentration at 90g/L.Mannitol obviously improved bulblet quality.The root growth and callus was promoted by increasing phosphorus concentration to a 1.5~2.0 fold of the standard MS,but bulblet enlargement was decreased.It was favorable for breaking bulblet dormancy by cold treatment at 5℃ for 45~60 days.When shoot sections from regenerated shoot clusters were cultured for bulblet formation,and the bulblets were cold treated for 40 days at 10~15℃,then the bulblets were storaged at 5℃ for 50 days,the percentage of bulblets sprouted with a stem with many leaves reached 37.8%.
