邻苯二甲酸丁苄酯(BBP)对雄性F1大鼠海马BDNF、CREST、SpinophilinmRNA表达的影响

【摘要】 目的:研究BBP对雄性F1大鼠海马神经行为发育相关基因mRNA表达的影响。方法:取PND1、7、21雄性仔鼠的海马,提取总RNA后逆转录获得cDNA,用荧光定量PCR法扩增,以β-actin为内参检测海马c-fos、CREST、BDNF、spinophilin mRNA表达。结果:(1)PND1雄性仔鼠海马基因mRNA表达改变:CREST mRNA在低剂量组表达下调(P<0.05),表达量为对照组的0.6倍,而在中、高剂量组表达上调(P<0.05),分别为对照组的1.5倍和1.6倍;spinophilin mRNA仅在中剂量组的表达上调(P<0.05),为对照组的1.3倍。(2)PND7雄性仔鼠海马基因mRNA表达改变:CREST、BDNF、spinophilin mRNA表达在中、高剂量组均下调(P<0.05)。CREST mRNA在中、高剂量组表达量分别为对照组的0.56倍和0.49倍;BDNF mRNA在中、高剂量组表达量分别为对照组的0.69倍和0.77倍;spinophilin mRNA在中、高剂量组表达量均为对照组的0.36倍。(3)PND21雄性仔鼠海马各基因mRNA表达无明显改变(P>0.05)。与对照组比较,各处理组雄性仔鼠各时间点海马c-fos mRNA表达无明显改变(P>0.05)。结论:BBP对新生雄性仔鼠(PND1、PND7)海马基因(CREST、BDNF、spinophilin)mRNA表达有一定的影响;但出生后21天(PND21),各处理组雄性仔鼠海马三种基因表达却未见显著变化。

【Abstract】 Objective: To investigate the effects of Butyl benzyl phthalate (BBP) on the mRNA expressions of the genes in the hippocampus related to neurobehavioral development in the male offspring. Methods: The hippocampus of the male offspring were obtained at PND1, 7, and 21. After abstracting the total RNA, cDNA was obtained from reverse transcription. Real time PCR was introduced to detect the expression levels of mRNA in genes of c-fos, CREST, BDNF and spinophilin. Results: (1) Changes of mRNA-expression of genes in hippocampus at PND1: compared to the control group, the expression of CREST mRNA was down-regulated (P<0.05) in the low-dose group (0.6 fold) and up-regulated (P<0.05) in the mid- and high-dose groups (1.5 and 1.6 fold). The expression of spinophilin mRNA was up-regulated (P<0.05) in the mid-group (1.3 fold). (2) Changes of mRNA- expression of genes in hippocampus at PND7: the mRNA expression levels of CREST, BDNF and spinophilin were extremely lower than the control group in the mid- and high-dose groups, with CREST 0.56 and 0.49 fold, BDNF 0.69 and 0.77 fold, spinophilin 0.36 and 0.36 fold in the mid- and high-dose group respectively.(3) The expressions of mRNA of the selected genes in hippocampus at PND21 were not obviously changed in any BBP-treated groups (P>0.05). The expressions of c-fos mRNA in all BBP-treated groups were not changed in any time point. Conclusion: BBP demonstrated some effects on mRNA expressions of the genes (CREST, BDNF, spinophilin) in the hippocampus of the male offsprings at PND1 and PND7 but not at PND21.
