以改性血红蛋白作为纳米生物催化剂酶法测定过氧乙酸

【摘要】 用牛血红蛋白(BHb)和交联血红蛋白(CHb)代替辣根过氧化物酶(HRP)作为纳米生物催化剂,硼酸盐缓冲液存在时,过氧乙酸(PAcA)和苯酚、4-氨基安替比林发生Trinder反应。考察了pH值、时间和温度对反应的影响。通过正交试验确定了反应的最佳条件。过氧乙酸以喷雾法和熏蒸法进行空气消毒,用气体吸收管现场采样,以酶催化分光光度法测定室内环境中的微量过氧乙酸消毒剂的含量。测量过氧化物的工作曲线的线性回归方程为A=0.0607+0.525c(μmol.L-1),线性相关系数ρ>0.994,溶液组分(PAcA+H2O2)的检出浓度为17.346μmol.L-1,对应25mL过氧化物检出限为43.365nmol,过氧乙酸的检出限0.330μg。

【Abstract】 Trinder coloring reaction of peracetic acid, phenol and 4-AAP, existing borate buffer solution, created the quinone dye matter which bull hemoglobin (BHb) or crosslinked hemoglobin (CHb), in place of horseradish peroxidase (HRP), was used as enzymy-like nanocatalyst in. The effects of pH、time and temperature to reaction were researched. The optimal temperature、time、the amount of phenol and value of pH were individual 50℃、50min、1.5mL and 7.73 by ort hogonal experimental design. The method of sprayer and fumigating method can be used when peracetic acid is applied as air disinfector. Different concentrations of micro dosage of indoor peracetic acid absorbed by gas absorption tube were measured by biocatalytic spectrophotometry. Equation of linear regression with workplot of determination of microscale peroxide was A=0.0607+0.525c (μmol.L-1) and linear regression coefficientρ> 0.994 . The limit of detection was 17.346 μmol.L-1,to 25 mL solution component, corresponding limits of detections to peroxide disinfector and to peracetic acid were 43.365 nmol and 0.330μg.
