RP-HPLC测定杜仲叶中京尼平苷酸、绿原酸、京尼平苷的含量
The Determination of Geniposidic Acid, Chlorogenic Acid and Geniposide in Folium Cortex Eucommia by RP-HPLC

【摘要】 目的建立反相高效液相色谱法同时测定杜仲叶中京尼平苷酸、绿原酸、京尼平苷含量的方法。方法色谱条件为Diamonsil-C18柱,甲醇-水-冰醋酸(17:83:1.5,PH=2.70)为流动相;流速1.0ml/min;检测波长为237nm。结果京尼平苷酸在0.36~3.60μg范围内线形关系良好(r=0.9999),平均加样回收率99.57%,RSD=1.49%(n=6);绿原酸在1.92~9.60μg范围内线形关系良好(r=0.9999),平均加样回收率100.21%,RSD=1.53%(n=6);京尼平苷在0.26~2.60μg范围内线形关系良好(r=0.9997),平均加样回收率98.97%,RSD=1.91%(n=6);结论3种成分的峰分离效果好,其测定方法操作简便、准确、重现性良好,可作为杜仲叶中京尼平苷酸、绿原酸、京尼平苷的质量控制方法。
【Abstract】 Objective: To establish a new RP-HPLC method for simultaneous determination of geniposidic acid(GPA), chlorogenic acid(CA) and geniposide(GP) in Folium Cortex Eucommia. Methods: The chromatographic condition was Diamonsil-C18 column, the mobile phase consisting of CH3OH-H2O-HAc(17:83:1.5,PH=2.70);the flow rate was 1.0 ml/min;the detection wavelength was 237nm.Results : The standard geniposidic acid showed a good linear correlation within the concentration of 0.36 ~3.60μg,(r=0.9999),the average recovery of the sample was 99.57%,RSD=1.49%(n=6);the standard chlorogenic acid showed a good linear correlation within the concentration of 1.92 ~ 9.60μg,(r=0.9999),the average recovery of the sample was 100.21%,RSD=1.53%(n=6);the standard geniposide(showed a good linear correlation within the concentration of 0.26 ~2.60μg,(r=0.9997),the average recovery of the sample was 98.97%,RSD=1.91%(n=6).Conclusion: The peaks of the three components were separated effectly,the method is simple,accurate and reproducible and can be used for the quality control of geniposidic acid(GPA), chlorogenic acid(CA) and geniposide(GP) in Folium Cortex Eucommia.
