银狐着丝粒卫星DNA序列的克隆及序列分析

【摘要】 本实验利用限制性内切酶Sac I酶切银狐基因组DNA,经1.0%凝胶电泳,回收特异性亮带,连接pUC18载体,转化DH5 α大肠杆菌,随机挑取10个阳性克隆测序采用DNAMAN 和BLAST进行序列分析, 其中9个克隆为银狐着丝粒卫星DNA重复序列,长度为741bp与 743bp之间,序列之间的同源性在90%与97%之间,经BLAST检索,这些重复序列与已报道的犬着丝粒卫星DNA重复序列同源性为80%左右。

【Abstract】 The prominent 750bp ethidium-bromide-stained bands was recovered d by electrophoresis after restriction enzyme digestion by Sac I. The fragments were introduced into DH5 a of Escherichia coli by pUC18 vector, the recombinant plasmid was identified by PCR, then ten recombinant clones was sequenced. Sequence analysis revealed that the nine sequences had 80% homolony to known dog satellite DNA sequence and the fragment was from 741bp to 743bp, the homology among the mine sequences is from 90% to 97%. By this experiment, it can indicate these sequences are the silver fox’s satellite DNA sequence.
