猪跨膜整合蛋白2B(ITM2B)基因内含子3中两个SNP位点与猪胴体和肉质形状的相关性分析(英文)

【摘要】 本研究是为了寻找跨膜整合蛋白2B(ITM2B)基因内存在的多态性位点,并初步探讨该基因内存在的多态性位点与胴体和肉质性状的相关性。利用两对引物,成功扩增出了一个2036 bp的片段。该片段包含了完整的外显子4、内含子3、内含子4和外显子3、外显子5的部分序列。比较中外猪种中的序列差异,在内含子3中存在2个SNP位点。其中C168T用PCR-HaeⅢ-RFLP方法检测,而A1125G则用 PCR-TaqⅠ-RFLP方法检测。这两个SNP位点在大白猪×梅山猪资源家系的多态性分析表明,C168T与瘦肉率(p<0.01),失水率(p<0.01),内脂重(p<0.01),花油(p<0.05),皮重(p<0.05),皮率(p<0.05),皮厚(p<0.05)存在显著或极显著相关;而A1125G与眼肌面积(p<0.01),胴体长(至第一肋骨)(p<0.05), 骨骼肌内脂肪(p<0.01)和骨骼肌水分含量(p<0.01)存在显著相关。对于这些初步的结果,还需要在更大的群体中和该基因的功能方面做深入研究,以验证这两个SNP位点与猪胴体性状和肉质性状的确切相关性。

【Abstract】 The study was aimed at investigating polymorphisms of the porcine integral membrane protein 2B (ITM2B) gene which is differentially expressed between parental purebreds of Large White and Landrace and their F1 hybrids Large White×Landrace, detecting by classical mRNA differential display technique. A 2036-bp fragment corresponding to partial exon 3 and 5, complete exon 4, intron 3 and 4 was amplified with two primer pairs. Sequence analysis revealed two SNPs in intron 3. The C168T mutation was detected by PCR-HaeⅢ-RFLP protocol whereas the A1 125G was detected by PCR-TaqⅠ-RFLP protocol. Polymorphism analysis in a pig resource family showed that significant associations were observed for ITM2B C168T polymorphic site with lean meat percentage (p<0.01), drip loss rate (p<0.01), internal fat weight (p<0.01), caul fat weight (p<0.05), skin weight (p<0.05), skin percentage (p<0.05), skin thickness (p<0.05), and Al 125G were significantly associated with lion eye height (p<0.01), carcass length to 1st rib (p<0.05), intramuscular fat of longissimus dorsi (P<0.01), water moisture of longissimus dorsi (p<0.01). Further studies on the specific gene function are needed to confirm these preliminary results.
