响应面法优化宇佐美曲霉产菊粉酶发酵条件
Optimization of inulinase production by Aspergillus usamii with Response Surface Methodology

【摘要】 对宇佐美曲霉产菊粉酶发酵条件进行了研究。通过单因素实验初步确定了菊芋汁为碳源,蛋白胨为有机氮源,(NH4)2HPO4为无机氮源。采用中心组合设计和响应法对宇佐美曲霉培养条件进行优化,得到优化培养基为:蛋白胨22.14 g/L,菊芋汁117.28 mL,(NH4)2HPO43.94 g/L,NaCl5.0 g/L,MgSO40.50 g/L和 MnSO40.10 g/L。在上述优化培养条件下,宇佐美曲霉产菊粉酶的酶活为11.02 U/mL。
【Abstract】 The optimal culture conditions of Aspergillus usamii which produce of inulinase was studied . Jerusalem artichoke juice was the sole carbon source. The peptone and (NH4)2HPO4 as nitrogen sources were found to be most suitable ones for inulinase production by single factor experiment. The optimal conditions were obtained by central composite designs and response surface analysis as follows: peptone 22.14 g/L, Jerusalem artichoke 117.28mL, (NH4)2HPO43.94 g/L, NaCl5.0 g/L, MgSO40.50 g/L, MnSO40.10 g/L. The enzyme activity was 11.02U/mL in the optimized condition.
