胃癌组织中RUNX3、P53基因表达与其临床生物学特性的研究

【摘要】 目的:探讨RUNX3和P53基因mRNA在胃癌组织与正常胃粘膜组织中的表达差异和其与临床病理的相关关系。方法:采用逆转录-多聚合酶链反应(RT-PCR)技术对临床80例胃癌新鲜标本组织及肿瘤周围正常组织粘膜中 RUNX3和P53基因的表达进行检测,对基因弱表达与各临床病理参数的关系进行分析。结果:R T—P C R结果显示 RUNX3mRNA在25.00%(20/80)阳性表达的胃癌组织中的光密度比值(0.5719%±0.1013%)明显低于在78.75%(63/ 80)阳性表达的正常粘膜中的光密度比值(0.8297%± 0.2912%),两组比较差异性显著(t=6.1435 p=0.001), P53mRNA在27.50%(22/80)阳性表达的胃癌组织中的光密度比值(0.6133%±0.0978%)明显低于在88.75%(71/ 80)阳性表达的正常粘膜中的光密度比值(0.8305%± 0.1895%),两组比较差异性显著(t=5.9672 p=0.001), RUNX3基因在肿瘤组织中表达与肿瘤分化程度和淋巴结转移有关,RUNX3和P53 mRNA在胃癌组织中表达成正相关 (r=0.9840 P=0.0001)。结论:RUNX3和P53mRNA在胃癌组织中的表达量明显低于正常组织并且两者表达成正相关。 RUNX3和P53基因的表达异常与胃癌的发生发展过程密切相关并且可能在胃癌的侵袭和转移过程中起重要作用。

【Abstract】 Objective: Exploring the expression of RUNX3 gene mRNA and P53 gene mRNA in human gastric carcinoma and surrounding normal tissue and the correlations with clinicopathological parameters. Methods: RT-PCR were used to detect expression of RUNX3 gene mRNA and P53 gene mRNA in 80 cases of human gastric carcinoma and surrounding normal tissue. Result: Expression of RUNX3 gene were found in the 25% (20/80)of cases of human gastric carcinoma showed significantly lower (0.5719% ± 0.1013% ) than 78.75% (63/80) of cases of surrouding normal tissue( 0.8297v ± 0.2912% ) (t=6.1435 p=0.001), Expression of P53 gene were found in the 27. 50% (22/80)of cases of human gastric carcinoma showed significantly lower (0.6133% ± 0.0978% ) than 88.75% (71/80) of cases of surrouding normal tissue (0.8305% ± 0.1895% ) (t=5.9672 p=0.001), There was a significant correlation between RUNX3 gene and P53 gene expressed in the gastric carcinoma(P<0.05). Conclusion: Lower expression of RUNX3 gene mRNA and P53 gene mRNA in the gasric carcinoma and high correlation between them. Abnormal expression of RUNX3 gene and P53 gene probably played an important role in the tumorigenesis and development of gastric carcinoma.
