中药土大黄煎剂及含药血清对食管癌细胞增殖反应的影响

【摘要】 目的:探讨中药土大黄煎剂及其含药血清的抗肿瘤及免疫调节作用。方法:以传统方法用蒸馏水浸泡土大黄干燥的根茎,煮沸后改文火继续30分钟,取上清过滤制备成A、 B、C高、中、低3组不同剂量(分别含土大黄生药为100、50 和10mg/ml)的煎剂,并用其制备了大鼠的含药血清。根据临床常用剂量,结合小鼠急性毒理试验结果使土大黄煎剂为原液,设定了9种(10、5、1、0.5、0.25、0.1、0.05、 0.025和0.01 mg/ml)不同浓度的土大黄煎剂。以MTT 法测定比较土大黄煎剂及其含药血清对食管癌细胞和食管癌患者外周血淋巴细胞的抑制作用。结果:土大黄煎剂在含生药量0.1 mg/ml时对食管癌细胞株有较明显的抑制作用,抑制率为51.9%;对食管癌患者癌细胞抑制作用相对较弱,抑制率为37.9%;对外周血淋巴细胞增殖抑制率为30.1%。该浓度是体外实验的最强作用浓度,随着煎剂浓度的增加或减低,其对各类细胞增殖的抑制率均降低。发现煎剂浓度为5 mg/ml 和10mg/ml时,对外周血淋巴细胞的抑制率是负值,说明反而有促进增殖作用;本实验制备的A、B、C三组含药血清对细胞增殖均有抑制作用。A、B两组含药血清对传代的食管癌细胞株有明显抑制作用,抑制率分别为36.7%和31.0%,而 C组含药血清抑制率则很低为6.9%,A、B两组与C组之间相比较差异均有统计学意义(P<0.01)。A组含药血清对外周血淋巴细胞抑制作用为33.0%,与正常大鼠血清(阴性对照) 和化疗药制备的含药血清(阳性对照)作用相比,差异均有统计学意义(P<0.05)。结论:土大黄煎剂及其含药血清均能抑制食管癌细胞增殖,作用与剂量和浓度有关;土大黄煎剂及其含药血清有抗肿瘤作用。不同剂量的煎剂对淋巴细胞作用明显不同,煎剂在较低的剂量时可抑制外周血淋巴细胞增殖, 而在高剂量时则促进外周血淋巴细胞增殖。显示出七大黄对具有免疫功能的淋巴细胞可能有双相调节作用,但其剂量用法及临床意义有待进一步探讨。土大黄煎剂对实体瘤原代食管癌细胞和传代的食管癌细胞的抑制作用不尽相同,可能与其在体内具有免疫调节作用有关。
【Abstract】 Objective: This study was designed to elucidate Rhubarb and drug serum anti-tumor and Immune modulation function. Methods: The stem of Rhubarb dipped into distilled water and seethed. It was prepared for three variety doses A、 B、 C group(100mg/ml、 50 mg/ ml and 10 mg/ml)to observe its inhibition effect for esoph-ageal carcinoma cell and peripheral blood lymphocytes (PBL) of patients by MTT assay. Results: There were different inhibition effects after variety dose drug and drug serum. Esophageal carcinoma cell was strong inhibited proliferation when Rhubarb was O.lmg/ml. Inhibition rate was 51.9%; while, it weak effects the esophageal carcinoma cell of patients. Inhibition rate was 37.9%. It gentle effects PBL of patients. Inhibition rate was 30.1%. The proliferation of PBL was induced when 5mg/ml and 10mg/ ml of Rhubarb. The inhibitory rates are 36.7%、 31.0% of the A and B group serum on carcinoma cell TE13 was much higher than that of the C group serum (P<0.01). The inhibition rate (33.0%) of the A group serum for PBL was significantly different compared with negative and positive control (P <0.05). Conclusion: Rhubarb and its drug serum inhibit esophageal carcinoma cell. It suggests rhubarb and its drug serum have anticancer and immune modulation effect Effect is related to rhubarb concentrate.
