RASSF1A基因在乳腺癌组织中的表达及临床病理意义

【摘要】 目的:探讨RASSF1A基因在乳腺癌组织中的表达以及与乳腺癌临床病理特征的关系。方法:采用逆转录聚合酶链反应(RT-PCR)技术检测49例乳腺癌组织及癌旁非肿瘤组织中RASSF1A基因的表达。应用条带密度分析软件,定量分析RT-PCR产物电泳带密度。结果:49例癌旁非肿瘤组织中RASSF1A全部表达,RASSF1A校正值 (RASSF1A/β-actin)为0.6902±0.1179,49例乳腺癌组织中仅19例RASSF1A表达,校正值为0.1833±0.2441, 癌组织与癌旁非肿瘤组织中RASSF1A基因表达差异有显著性(p< 0.01)。随着临床分期的增高,RASSF1A基因阳性表达率降低(r=-0.447),病理组织学分级增加,RASSF1A基因呈现低表达(r=-0.402),c—erbB-2蛋白表达阳性伴随 RASSF1A基因的低表达(r=-0.613),即RASSF1A基因高表达与临床分期、淋巴结转移、病理组织学分级、c— erbB-2蛋白表达呈负相关(P<0.01)。而与患者年龄(r= 0.196)、病理组织学类型(r=0.100)及其雌、孕激素受体 (ER/PR,r=-0.082,r=-0.173)状况无关(P>0.05)。结论:乳腺癌旁非肿瘤组织中RASSF1A基因表达高于癌组织,随着临床分期的增高、病理组织学分级的上升以及c— erbB-2蛋白的高表达,RASSF1A基因表达率降低。提示, RASSF1A基因丢失与乳腺癌的发生、发展及乳腺肿瘤较差的生物学特性相关。

【Abstract】 Objective: To study the expression of RASSFIA gene in breast cancer and its clinicopathologic characteristics. Methods: With β -actin gene as reference, the RASSFIA was monitored in 49 specimens of breast cancer tissue and paired non-tumorous tissues by reverse transcription-polymerase chain reaction (RT-PCR). Results: The RASSFIA expression was significantly upregulated in whole paired non-tumorous tissues with 0.6902 ± 0.1179, which was only expressed in 19 cases of breast cancer tissues with 0.1833 ± 0.2441. The difference of RASSF1A expression between cancer and paired non-tumorous tissues was significant (p<0.01). RASSFIA expression in breast cancer was negative correlation with its TNM stage, lymph node metastasis, histological grade, c-erbB-2 protein overexpression (p<0.01), but no relation- ship with age, pathologic classification and ER, PR (p> 0.05). Conclusion: The level of RASSF1A expression is obviously higher in the paired non-tumorous tissues than in breast cancer. Lower expression of RASSFIA gene are followed by higher TNM stage, lymph node metastasis, his-tological grade and c-erbB-2 protein overexpression. This supports the notion that absent of RASSFIA gene may play a crucial role in the carcinogenesis, development and worse biologic characteristics of breast cancer.
