应用RT-PCR检测乳腺癌患者骨髓MUC1mRNA的临床意义

【摘要】 目的:探讨检测乳腺癌患者骨髓中MUC1mRNA 的表达及其临床意义。方法:应用RT-PCR技术,同时对照检测75例乳腺癌患者、15例乳腺良性病变患者和8例健康人骨髓中的MUC1mRNA表达,并分析MUC1mRNA表达与临床病理因素和相关分子指标(Ki67、p53和VEGF) 的关系。结果: 75例乳腺癌患者中,32例检测出 MUC1mRNA阳性表达,阳性表达率为42.7%(32/75)。乳腺良性病变患者和健康人骨髓中未检测到MUC1mRNA表达。MUC1mRNA阳性表达与乳腺癌临床分期、腋窝淋巴结转移和ER状况有关(P<0.05),与年龄、肿瘤大小和PR 表达状况无统计学意义(P>0.05),与乳腺癌组织中Ki67、 p53和VEGF表达均无相关。结论:MUC1mRNA可作为检测乳腺癌患者骨髓微转移的分子指标之一。骨髓微转移的检测为乳腺癌患者的治疗和预后判断提供帮助。

【Abstract】 Objective: This study was designed to evaluate expression and clinical significance of the mucinl (MUC1) mRNA in bone marrow of patients with breast cancer. Methods: Expression of MUC1 mRNA was detected by reverse transcriptase polymerase chain reaction (RT-PCR) in the bone marrow aspiration from 75 breast cancer patients, with samples of 15 benign breast lesion and 8 healthy volunteers as control. We analyzed the pos- sible correlation of MUClmRNA expression with clinico-pathologic parameters and related molecular markers. Results: The positive expression of MUClmRNA was detected by RT-PCR in 32 of 75 patients with breast cancer. The positive rate was 42.7%. No MUClmRNA expression was found in the bone marrow aspiration of patients with benign breast lesion and healthy volunteers. The MUClmRNA expression was positively correlated with clinical stage , axillary nodal involvement and ER status ( P<0.05 ), but was not related to age, tumor size, and PR status(P>0.05). In addition, positive rate of MUClmRNA was not correlated with expression of Ki67, VEGF and p53 in tissues of breast cancer. Conclusion: MUClmRNA may be used as a molecular marker to detect bone marrow microwetastasis( BMM) in patients with breast cancer. The results of detecting BMM may have an impact upon therapeutic recommendations and prognostic judgement in breast cancer patients.
