nm23、p53基因突变与贲门癌癌生物学特性的关系

【摘要】 目的:检测贲门癌组织中nm23和p53基因外显子突变情况,寻找有助于准确预测局部肿瘤进展、分型、预后情况的临床参照指标。方法:应用PCR—SSCP检测21例贲门癌术后新鲜标本组织中nm23及p53基因外显子突变情况,并与贲门癌的病理及临床指标进行相关性研究。结果: nm23表达及p53基因突变率分别为71.4%及48.86%。nm23 和或p53基因突变与临床分期呈正相关(P<0.01)。结论: nm23和/或p53基因突变与贲门癌临床分期相关。nm23基因表达及p53基因突变在判断贲门癌细胞增殖、分化程度、恶性进度及预后转归方面有一定价值,可作为判断贲门癌预后的参考指标,指导高危患者的进一步治疗。

【Abstract】 Objective: Our study aimed to investigate whether nm23 expression and p53 gene mutation in the primary Cardiocarcinoma correlated with tumor grade stage recurrence metastasis and patient survival time. Methods: RT-PCR and PCR-SSCP is used to test the expression in nm23, test the mutation in p53 of the samples. 21 cases paraffinem beded samples were admitted. Results: The gene expression rate of nm23 were 71.4% and the gene mutation rate of p53 were 48.86%. All expression and mutation happened in the low grade tumor. In this study, nm23 gene express and p53 gene mutation were correlated with histological grade and clinical stage and closely correlated. Conclusion: We found that Nm23 and p53 play an important role in the development and progression of Cardiocarcinoma. nm23 and p53 appear to be associated with the proliferation and progression of Cardiocarcinoma. Both of them can be used as prognostic factors in Cardiocarcinoma.
