茶多酚指纹图谱的HPLC和LC-MS/MS研究

【摘要】 目的:建立茶多酚的HPLC指纹图谱标准。方法:采用ODS柱,流动相A:甲醇-0.1%甲酸溶液(10:90)与流动相B:甲醇-乙腈-0.1%甲酸溶液(50:30:20),进行梯度洗脱,检测波长280nm。并对主要成分峰进行LC-MS/MS鉴定。结果:茶多酚HPLC指纹图谱中有11个特征成分峰,经对照品对照及液质联用鉴定了其中9个成分,分别为EC、EGC、ECG、EGCG等。结论:HPLC指纹图谱可以对茶多酚中主要特征有效成分进行准确的定性与定量测定,色谱系统稳定适用性良好;通过指纹对照品对照测定可以准确控制茶多酚制剂质量。

【Abstract】 OBJECTIVE: To establish the fingerprint chromatogram standard of Tea Polyphenols by HPLC. METHODS: Separation was performed on ODS column with gradient elution by the mobile phase consisting of methanol-0.1% formic acid(10:90) and methanol-acetonitrile-0.1% formic acid(50:30:20). The detection wavelength was set at 280 nm. And most of chromatogram peaks were identified by LC-MS/MS RESULTS: There were 11 common peaks in fingerprint chromatograms and 9 of them were identified with reference and LC-MS/MS, which were EC, EGC, ECG, EGCG and etc. CONCLUSION: The method is accurate in identification and determination of compounds in tea polyphenols. The chromatographic system is stable and applicable.
