中药益母草的rDNA ITS序列分析

【摘要】 目的:分析ITS片段在中药益母草DNA分子鉴别中的重要作用,也为中药材基因芯片的研究奠定一定的基础。方法:对国内不同产地容易与中药益母草混淆的4个种分别进行rDNA ITS序列的PCR扩增和测序,并运用CLUSTRAL X和MEGA软件进行分析。结果:测得这4种植物的片段包括ITS1、5.8S和ITS2全长序列以及18S、26S部分序列,益母草及其易混淆的3种植物ITS1、ITS2序列的差异性分别为7.2%~18.8%和14.2%~27%。根据ITS序列特征所构建的系统树与传统分类有所不同。结论:rDNA ITS序列特征可作为益母草种间鉴别的较有效分子标记。

【Abstract】 Objective: To analyze the important utility of ITS sequences in molecular authentication of Leonurus artemisia(Lour.)S.Y.Hu, and establish foundation for the research of DNA chip of Traditional Chinese Medicine. Methods: The rDNA ITS regions of L.artemisia and its adulterant species were amplified and sequenced. CLUSTRAL X and MEGA softwares were applied for their analysis. Results: The internal transcribed spacers (ITS) including ITS1, 5.8S, ITS2, and partial 18S and 26S were determined. The range of diversity among L.artemisia and its adulterant species was 7.2%~18.8% and 14.2%~27% based on ITS1 and ITS2 respectively. The dendrograms based on rDNA ITS sequence data were different from the result from morphology. Conclusion: The rDNA ITS sequence can be used to be an efficient identification marker of L.artemisia and its adulterants. And it needs further study on the phylogenesis of Leonurus plants.
