注射用姜黄素冻干脂质体的质量评价

【摘要】 目的:建立自制姜黄素冻干脂质体的质量标准。方法:采用Zetasizer粒径测定仪测定粒径;采用反相高效液相色谱法(RP-HPLC)测定姜黄素冻干脂质体的含量;采用离心超滤法测定脂质体的包封率。结果:姜黄素冻干脂质体粒径大小均匀,平均粒径0.732μm;RP-HPLC法测定,在5~80μg·ml-1浓度范围内线性关系良好,r=0.9997;日内和日间精密度均良好(RSD<2%);高、中、低3个浓度的回收率分别为103.5%,98.5%,102.0%;最低检测限8ng;脂质体包封率达97.45%。结论:本评价方法准确可靠,重现性好、操作简便、专属性强,符合质量测定要求。
【Abstract】 Objective: To establish a quality standard for curcumin freeze-drying liposomes. Methods: Zetasizer was used to study the size of liposomes. A RP-HPLC method was developed to determine the drug content. The entrapment efficiency was determined with centrifugal ultrafilter. Results: The freeze-drying liposomes were homogeneous and the average size was 0.732μm.The standard curve was linear over the range of 5~80μg?ml-1, r=0.9997,The intra-day and inter-day RSD was less than 2.0 % .The recoveries of the high, middle and low concentration were 103.5%,98.5%,102.0% respectively, the detection limit was 8 ng, and the entrapment efficiency was 97.45%.Conclusion:This method is simple, accurate and practical for evaluating the quality of curcumin freeze-drying liposomes.
