酶联免疫吸附法快速检测鳗弧菌

【摘要】 鳗鲡弧菌病是鳗鲡养殖中主要的疾病之一,危害大,死亡率高;同时鳗弧菌 (V.aguillarum)也是许多鱼类易感的致病菌,常引起鱼类大量死亡。为此,我们制备了鳗弧菌E3-11菌株的兔抗血清,并应用间接酶联免疫吸附法,建立了一种新的快速诊断鳗鲡弧菌病的方法。该方法能检出1.5×105个/mL浓度的E3-11菌株,对患病鱼体的组织培养物鳗弧菌进行检测,呈现明显的阳性反应。对嗜水气单胞菌(A.hy- drophila),柱状纤维粘细菌(C.columnaris),非01霍乱弧菌(Non-01)等几种鳗鲡养殖中主要致病菌不发生交叉反应,同时该反应过程可在较短时间内完成。该方法具有敏感性高、特异性强、速度快、实用方便等特点,可应用于鳗弧菌病的快速检测和诊断。

【Abstract】 Eel vibriosis is one of the main diseases detected during the eel culture with great risk and high mortality. Meanwhile, Vibrio anguillarum is also the sensitive pathogenic bacteria for many fishes,causing mass death among fishes. Therefore, we prepared rabbit antiserum for E3-11 strain of V. anguillarum,applied indirect ELISA and established new method for the rapid diagnosis of eel vibiosis. The method was able to detect E3-11 strain at the concentration of 1.5×105 cells/mL and demonstrated obvious positive reaction when checked on V. anguillarum in the tissue culture of affected fishes. The method did not have cross reaction to several pathogenic bacteria during the eel culture,e. g. , Aeromonas hydrophila , Cytophaga columnaris, Non-01 comma bacillus ,and the reaction would be completed in a short time. In a word this method has high sensibility,high singularity and high speed as well as easy usage and can be utilized in rapid detection diagnosis of eel vibiosis.
