龋损及牙髓炎症时人牙髓组织中LPS信号受体表达研究

【摘要】 目的:研究人正常牙髓组织、龋病牙髓组织和牙髓炎牙髓组织中LPS信号受体CD14、TLR4的表达特点,探讨深龋和牙髓炎中LPS的信号转导途径。方法:应用直接免疫荧光际记法,采用流式细胞术检测人牙髓炎症组织中炎症细胞的CD14、TLR4 的平均荧光表达强度。结果:正常牙髓组织中CD14、TLR4平均荧光表达强度无显著性差异(P>0.05),龋病牙髓组织中CD14平均荧光表达强度增高(P<0.05),牙髓炎牙髓组织中CD14平均荧光表达强度显著增高(P<0.05),龋病牙髓组织和牙髓炎牙髓组织中TLR4平均荧光表达强度无显著性差异(P>0.05)。结论:龋病牙髓组织和牙髓炎牙髓组织中CD14平均荧光表达强度均增高提示LPS可能通过CD14信号受体在牙体硬组织疾病中发挥作用,而且作用强度与炎症程度呈正相关。
【Abstract】 OBJECTIVE: To investigate the expression of LPS receptors-CD 14 and TLR4 in healthy, carious and pulpitical human dental pulp tissues and to explore the signal transduction pathway of LPS. METHODS: CD14, TLR4 expressing intensity in healthy, carious and pulpitical human dental pulp tissues were observed by flow cytometric detection in the form of the mean fluorescence intensity. RESULTS: CD14 and TLR4 were not detected in healthy pulp tissues. Compared with normal pulp tissues, the mean fluorescence intensity of CD14 were increased significantly in carious dental pulp tissues, and stronger in pulpitical dental pulp tissues. However, there were no TLR4 expression in carious and pulpitical human dental pulp tissues. CONCLUSION: This study-revealed a significant increase in the expression of CD14 in carious and pulpitical human dental pulp tissues. LPS may plays important roles in the process of caries and pulpitis, and the expressing intensity increases as the inflammatory grows on.
