【摘要】 <正>目的:制备羟基喜树碱(HCPT)/聚乙二醇-聚谷氨酸苄酯(PEG-PBLG)纳米载药微球进行体外抑制Tca8113细胞作用研究。方法:透析法制备HCPT/PEG-PBLG纳米微球;电镜检测纳米微球结构表征,紫外分光光度仪测定样品吸光度后计算包封率和载药率。HCPT/PEG-PBLG纳米微球对Tca8113 细胞体外抑制实验:观察细胞形态学改变,MTT检测细胞毒作用。流式细胞仪、DNA凝胶电泳检测

【Abstract】 Objective: To prepare HCPT/PEG-PBLG nanoparticles and investigate the inhibiting effects to Tca8113 cell. Methods: Synthesized PEG-PBLG nanoparticles loaded closed ring form HCPT by diafiltration methods. Measure the encapsulation efficiency and loading level of HCPT/PEG-PBLG by ultraviolet spectrophotometry. The structure & properties of the nanopar- tides including the size shape was inspected under Scan & Transmission Electron Microscopy. Inhibitation experiment of HCPT/PEG-PBLG nanoparticles to Tca8113 cells in vitro: The morphologic change, characteristics of growth and transmigration of the cell were studied under inverted phase contrast microscope and TEM. The cytotoxicity effect was evaluated by MTT assay and the cell cycle change and apoptosis were assessed with flow cytometry (FCM) and DNA gel electrophoresis. Results: PEG-PBLG nanoparticles had been successfully prepared, loading HCPT with 56.8% in encapsulation efficiency and 7.5% in loading level. Diameter of the nanoparticles was about 300 nm. Results of MTT assay showed that HCPT had obvious toxic action that cell growth inhibiting rate(GI) increased with concentration upgraded and time duration. HCPT/PEG-PBLG showed similar effectiveness but relatively delayed at check point to that of HCPT: GI value was less than HCPT at 48 hours (p=0.01) and equal to HCPT at 96 hours (no statistical significance was found) (p>0.05). Conclusion: HCPT, appeared to fairly restrained tumor growth of Tca8113 cell in vitro. HCPT delayed cell cycle in S phase, inducing apoptosis. HCPT /PEG-PBLG nanoparticles slowed down the drug release, so, compared wih HCPT, it showed milder reactions.
