【摘要】 <正>目的:体外观察顺铂诱导舌鳞癌细胞株(Tca8113)凋亡的作用,同时检测在此过程中凋亡蛋白抑制因子Survivin mkRNA表达和蛋白表达的变化,为探讨舌鳞癌的化疗药物耐药机制提供理沦基础。方法:人 Tca8113细胞株进行传代培养;MTT法检测顺铂以不同浓度和不同时间对Tca8113细胞生长的抑制作用: 通过RT-PCR检测凋亡过程中Survivin基因mRNA表达,免疫细胞化学观察Survivin基因的蛋白表达, 以及流式细胞仪(FCM)观察顺铂(DDP)诱导各组Tca8113细胞的凋亡。结果:顺铂可明显抑制Tca8113细
【Abstract】 Objective: To observe the induction of apoptosis of cisplatin(DDP) to oral squamous cell line (Tca8113) in vitro and study the role of survivin on the apoptosis of Tca8113 cells induced by cisplatin, so as to provide the basis for validating the mechanisms of chemotherapeutical drug resistance. Methods: The inhibitory effects of different doses of DDP on Tca8113 cells were assayed with MTT test. Apoptosis was determined by flow cytometry. The expression of survivin was detected by RT -PCR and immunocytochemistry. Results: Cisplatin obviously inhibited Tca8113 cell growth in a dose and time dependent manner. The apoptotic index showed the similar trend. Survivin expression was decreased with increasing of time and reached the lowest level at 24 hours after DDP treatment, then increased after that time. Conclusion: Cisplatin can effectively induce apoptosis in Tca8113 cells and the inhibition of survivin gene expression may play a critical role on Tca8113 cell apoptosis induced by cisplatin.
