【摘要】 <正>目的:研究诱导培养的SD大鼠骨髓基质细胞(bone marrow stromal cells,BMSCs)在新型纳米磷酸钙陶瓷支架材料上的粘附和生长。方法:体重180g SD大鼠2只,颈椎脱臼处死后取双侧股、胫骨,将骨髓冲入培养液诱导培养,经碱性磷酸酶(ALP)和Yon Kossa’s矿化沉积检测证实培养细胞其已具成骨细胞表型后,与多孔纳米磷酸钙陶瓷支架(实验组)及普通多孔磷酸钙陶瓷支架材料(对照组)体外复合培养, 扫描电镜观察比较实验组与对照组BMSCs在支架表面的贴附数量和形态;同时以不同浓度细胞悬液接
【Abstract】 OBJECTIVE To study the adhesion characteristic I of bone marrow stromal cells (BMSCs) that has been induced and proliferated on newly - designed porous calcium phosphate nano - bioceramics. METHODS BMSCs derived from SD rat bone marrow were induced and proliferated in vitro. After their osteogenic phenotypes were demonstrated, BMSCs were seeded onto prepared porous calcium phosphate nano - bioceramics and onto common calcium phosphate bioceramics as a Control ,Their adhesion situation were analyzed by scanning electron microscope . To investigate the initial optimal cell seeding density between newly - designed porous calcium phosphate nano - bioceramics by MTT automated coiormetric microassay method. RESULTS The osteoblastic phenotypes of differentiated BMSCs were demonstrated by the positive staining of mineralized node ,. ALP and Col I , the most appropriate seeding density between them was 1. 0 × 1067 cm2. The maximal number which MSC could adhere to decalcified bone is 1. 2× 107/cm3. CONCLUSIONS Bone marrow stromal cell can differentiate to osteoblast phenotype. The BMSCs were well adhered to new pattern porous calcium phosphate nano -bioceramics,
