口腔鳞癌中端粒酶活性和P53与Bcl-2的表达

【摘要】 <正>目的:测定口腔鳞癌组织端粒酶活性及P53和Bcl-2的表达,并探讨它们之间的关系。方法: 对31例口腔鳞癌新鲜活体组织分别用TRAP-银染法测定其端粒酶活性,用免疫组化S-P法检测 P53和Bcl-2的表达。结果:在87.1%(27/31)的口腔鳞癌组织中检测到端粒酶活性,同组病例P53
【Abstract】 Objective: To determine the expression of telomerase and the association between telomerase, p53 and bcl-2 in oral squamous cell carcinoma (OSCC). Methods: Thirty-one patient samples of squamous cell carcinoma samples were assayed using telomeric repeat amplification protocol (TRAP) method for detection of telomerase activity. The human osteosarcoma cell line, HOS cell, was used as a positive control and the lysis buffer, as a negative control. The expression of p53 and bcl-2 were detected by immunohistochemistry staining (S-P technique). Results: Twenty-seven (87.1%) of 31 OSCC samples were telomerase positive. P53 expression was detected in 15 (48.4%) of 31 tumor sepcimens by nuclear staining. Bcl-2 expression detected in 6 (19.4%) of 31 OSCC samples by plasma staining. In those telomerase positive samples, 55.6% (15/27) was p53 positive while 14.8% (4/27) bcl-2 positive. Conclusion: Telomerases may play a very important role in the development of OSCC. Telomerase activity might be associated with p53 expression, but not with bcl-2 in OSCC.
