【Abstract】 Objective: To observe the inhibitive effects of the angiogenesis inhibitor, TNP-470, on the growth and lung metastases of highly lung metastaic salivary adenoid cystic carcinoma (ACC-M) in nude mice, to investigate the mechanisms of TNP-470 against ACC-M in vivo, with the aim of clinical trial in the future. Methods: The nude mice were randomized into three treatment groups(15,30 and 60 mg/kg) and a control group. From the second week, TNP-470 was administered subcutaneously every day for two weeks. The mice were sacrificed at the seventh week after implantation. The lung metastasis rates and weights of mice detected. The numbers and area of metastases and ratio of area of metastases were analysed. The microvessel density (MVD), cell proliferation index (PI) and apoptosis index (AI) of the ACC-M lung metastases were observed. Results: Compared to the control, the lung metastasis rates, whte weightm, the numbers. the area of metastases, the ratio of area and the MVD of metastases were significantly reduced in mice treated with 30 and 60mg/kg TNP-470 (P<0.05). The AI of ACC-M cell was higher in the experimental groups (P<0.05). Conclusions: TNP-470 suppressed the growth and lung metastases of ACC-M in vivo. The inhibitive effects of 30 and 60mg/kg TNP-470 treated were better. We suggest that inhibiting angiogenesis, inducing apoptosis are the mechanisms of TNP-470 on the growth and metastases of ACC-M in nude mice.更多还原
