黄瓜原生质体培养与融合研究

【摘要】 黄瓜子叶原生质体来源的愈伤组织在1.0mg/L BA和0.05mg/L NAA的MS固体培养基上再生绿苗,经生根成为完整植株。10μg/ml诺丹明6G(R6G)处理15min或1.5mmol/L 碘乙酸(IOA)处理10min,均能有效地抑制黄瓜原生质体分裂。分别经R6G和IOA生理性失活的原生质体经PEG和高钙高pH法融合后可恢复分裂生长。由4个黄瓜种内组合得到了愈伤组织。部分愈伤组织分化了根,部分愈伤组织经酯酶同功酶分析确定为体细胞杂种。

【Abstract】 Plantlets were regenerated from calli which were originated from mesophyll protoplasts of Cucumis sativus and were cultured in solid MS medium supplemented with 1.0 mg/L BA and 0. 05 mg/L NAA. The protoplasts from several cultivated varieties and a semiwild variety were restained from division after they were treated either with rhodamine 6G at 10 μg/mL for 15 min or with iodoacetic acid at 1. 5 mmol/L for 10 min. However, the division potential can be resumed through fusion between protoplasts treated with either of the two metabolic inhibitors. Numerous calli were obtained from four kinds of introspecific combination of cucumber, some of which differentiated roots and/or were indentified as somatic hybrid tissues according to isozyrae analyses,
