芭蕾苹果微繁殖中抑制褐化的研究

【摘要】 本文对影响芭蕾苹果微繁殖中褐化的因素进行了研究。结果表明外植体在低温7℃,1/2 MS条件下培养5天可以抑制褐化,高盐浓度加重褐化,光强对褐化的影响差异不显著。培养基中附加抗坏血酸(VC)及柠檬酸(CA)可以减轻褐化,而加有活性炭(AC)及聚乙烯吡咯烷酮(PVP)的培养以及抗氧化剂溶液预处理外植体不能减轻褐化。对外植体母株枝条进行暗处理可以使褐化完全得到抑制。

【Abstract】 Several factors influencing phenol-induced browing of Ballerina apple were investigated by using shoot -tip explants cultured in vitro. Explants cultured with 1/2MS medium under 7℃ for 5 days produced low browning percentage and high surviving percentage. In any treatments, explant released more phenolic exudate on medium having high salt concentration. The browning percentage under light and dark were insignificantly different. Soaking the explants in water or ascobic/citric acid solution can’t prevent browning. Addition of 0. 25% activated charcoal(AC) or 0.1% polyvinylpyrrolidone(PVP) to the initiation medium can’t slow browning while 100/150mg/L ascorbic/citric acid(VC/CA)favoured establishment of Ballerina apple. Lowest incidence of browning and highest surviving percentage were obtained after 30 days of dark treatment for the branches of stock plants.
