高碘摄入对仔鼠小脑髓鞘蛋白影响的研究

【摘要】 目的:观察在不同碘摄入量下子代大鼠小脑中髓鞘碱性蛋白(MBP)mRNA及其蛋白表达水平,探讨高碘对小脑髓鞘发育的影响。方法:分别检测14日龄和28日龄仔鼠血清甲状腺激素TT4、小脑MBPmRNA和其蛋白的表达,应用 MIAS-2000型图像分析系统进行量化分析。结果:14日龄仔鼠的各项指标各高碘组与正常对照组差别均无统计学意义; 而28日龄100倍高碘组仔鼠的TT4水平低于正常对照组有统计学意义,MBPmRNA表达水平高于正常对照组有统计学意义,其余各高碘组与正常对照组差别均无统计学意义;髓板处MBP蛋白表达水平各高碘组与正常对照组差别均无统计学意义,髓鞘分枝处MBP蛋白表达水平各高碘组与正常对照组差别均有统计学意义。结论:碘过量影响了仔鼠小脑髓鞘分枝的发育,但程度较小,提示仔鼠对碘过量具有较高的耐受性。

【Abstract】 Objective To study the levels of myelin basic protein gene and protein expression in fetal rat subjected to different level of iodine intake. Methods Detect thyroid hormone TT4 in serum, MBP mRNA and protein expression in cerebellum of rat on postnatal 14 ( P14) and 28 ( P28 ) measured by MIAS-2000 Image Analysis System. Results No difference was found among HI and NI groups on P14. On P28 the level of TT4 in 100HI was lower than that in NI group, while the MBP mRNA expression in 100HI was higher than NI group, and there was no other difference a-mong 10HI, 50HI and NI groups. The expression of MBP protein in medulla in HI groups was not different with NI, but in ramification the expression of MBP protein in HI groups was lower than that in NI group. Conclusions The cerebellum myelination of rat is affected by iodine intake, but is weak, which illuminates fetal rat has a stronger tolerance to i-odine intake.
