苏云金杆菌抗虫基因cry IA转化果蔗的研究

【摘要】 以幼嫩叶鞘为外植体,对果蔗植株再生和遗传转化进行了研究。初步建立了高效的果蔗再生体系, 即幼嫩叶鞘为诱导愈伤组织的外植体,MS+2.4-D 2.0 mg/L 为诱导愈伤培养基,MS+2.4-D2.0 mg/ L+6-BA 1.0 mg/L 分化培养基,MS+IAA 2.0 mg/L 为生根培养基。同时确定了愈伤组织潮霉素筛选压为50 mg/L。利用农杆菌介导法将含有 cryIA 基因和 CPT I 基因的植物表达载体 pMG225导入果蔗组织,经潮霉素筛选、PCR 以及 Southern 杂交分析证实 cryIAc 基因已整合进果蔗基因组中。

【Abstract】 Plant regeneration and transformation from meristematic explants through callus induction were investi- gated in a case of fruit sugarcane cuhivar “Badila”.The optimal media were selected for plant regeneration by supplementing MS medium with different kinds of hormones:MS supplemented with 2.0 mg/L 2,4-D for pro- ducing callus;MS+2.4-D2.0 mg/L+6-BA 1 mg/L for differentiation;And MS+IAA 2.0 mg/L for inducing root.The optimal selective agent was 50 mg/L of hygB for callus and embryo culture.Plant expression vector pMG225 including cry IA gene and CPTI gene was transformed into fruit sugarcane tissues by Agrobacterium tu- mefaciens.PCR screening showed that some transgenic plants were positive,which indicated the foreign gene was seemed to integrate into fruit sugarcane genome.
