【摘要】 目的:了解近年来宁波市乙型流行性感冒病毒流行和抗原性的变异情况及种系分布.方法:采用狗肾传代细胞(MDCK)培养分离流感病毒,经血清学鉴定后提取病毒RNA, 经聚合酶链式反应扩增目的基因后进行乙型流感病毒的HA1区域基因片段的核苷酸序列测定.结果:2003年-2005年6月共分离到乙型流感病毒22株,血清学鉴定结果除2株为Victoda系的毒株以外,其余20株均为Yamagata种系.在时间的分布上2003及2004 年都有Yamagata和Victoria系的毒株出现.血凝素基因重链区(HA1)基因片段的核苷酸序列测定结果,两个系列的毒株都有一些小的变异,但Yamagata系的毒株变异更明显些. 2005年分离到的Yamagata系乙型流感与Shanghai/01/2000株相已经有7个氨基酸被替代, 它们的同源性只有97.6%.结论:虽然近年来宁波市流感病毒的主要流行椿一直以甲3型流感病毒为优势株,但乙型流感病毒也同时存在,虽然大多以散发存在为主,但在2004年底乙型流感病毒的Yamagata系曾出现了局部暴发流行,其抗原性有一定的漂移.更多还原

【Abstract】 Objective: Understand influenza B viruses circulating and antigenic drift and distributing since 2003 in Ningbo city. Method: Viruses were isolated with Madin-Darby canine kidney (MDCK) cell. Viral RNA was extracted from the viruses strain after serological identification and transcribed into cDNA by reverse transcriptase and amplified by PCR .Finally, the PCR product was purified and determined nucleotide sequence of HA1 gene of the influenza B viruses. Results: 22 influenza B viruses were obtained from 2003 to the June of 2005, including 2 of Victoria-lineage and 20 of Yamagata-lineage. On the distribution of time, both Victoria-lineage and Yamagata-lineage viruses were isolated in 2003 and 2004. The result of sequence analysis of the hemagglutinin (HA1) genes, there were several differ in amino acids of both Victoria and Yamagata lineages. Especially, Yamagata lineages viruses were more frequently, our Ningbo 2005 isolates differed by 7 amino acids from Shanghai/01/2000, the homology were 97.6%. Conclusion: Although the influenza A (H3N3) virus were most frequently in Ningbo during the previous 3 years, but influenza B were presence all the same with the quomodo sporadically mostly, furthermore, Yamagata lineages viruses even come forth outbreaks locally in the end of 2004, it’s antigenicity appreciably drifts.更多还原
