平肝熄风汤对大鼠脑出血后活化Caspase-3表达的影响
Effect of Pinggan Xifeng decoction on the expression of activated Caspase-3 after intracerebral hemorrhage in rat

【摘要】 目的观察大鼠脑出血后血肿周围神经元活化Caspase-3的表达情况、血肿变化、及平肝熄风汤的干预作用。方法用Ⅶ胶原酶脑内立体定位注射诱导大鼠脑出血模型,免疫组化检测血肿周围活化Caspase-3的表达,同时用直尺测量不同时间点大鼠血肿的最大直径。结果Ⅶ胶原酶造模后模型组血肿周围于6h出现阳性表达细胞,1d阳性细胞计数达高峰,3d 出现轻度下降,5d明显下降。与假手术组比较差异明显(P<0.01)。平肝熄风汤组与同时间点模型组比较:6h计数无明显差异,1d,3d,5d均存在明显差别(P<0.01).造模后血肿于1d达到最大,3d有所变小,5d变小明显;治疗组在1d与模型组比较血肿缩小明显(P<0.05),3d、5d 缩小更明显(p<0.01)。结论大鼠脑出血后血肿周围神经元活化Caspase-3表达明显上调;平肝熄风汤能下调活化Caspase-3的表达,阻止神经元的凋亡,同时促进血肿的吸收。
【Abstract】 objective to observe the effects of Pinggan Xifeng decoction(PGXF) on the expression of activated Caspase-3 in neuron around hematoma after intracerebral hemorrhage(ICH) in rat, and on the change of hematoma. Methods Ⅻ type collagenase was stereotaxically injected into the globas pallidus of rats to build the ICH model, immunohistochemical methods was used to exam the expression of activated Caspase-3 after ICH in rat .and ruler was used to measure the biggest diameter of rat hematoma at different time . Results the positive cells appeared around the hematoma at 6 hours after operation , and peaked to the top at I day , then descend lightly at 3 days , it descend distinctly at 5 days . compared with the fake group, it varied evidently(p<0.01). the number of positive cells of the PGXFD group was obviously smaller than that of the model group at 1d、 3d、 5d(p<0.01),but not at 6h.the hematoma swelled to the biggist at 1d , deflated lightly at 3d,and became more small at 3d. the hematoma of treatment group deflated smaller than that of model group at Id and 3d(p<0.05),it deflated more obviously at 5d(p<0.01). Conclusion the activated Caspase-3 increased obviously in the neuron around the hematoma after ICH; PGXFD can markedly depress the expression of activated Caspase-3, prohibit neuronal apoptosis, and accelerated the absorbing of the hematoma.
