钟基因Clock及钟相关基因在外周血淋巴细胞中的表达规律

【摘要】 目的探讨生物钟基因Clock及相关褪黑素受体基因(mt1和mt2)在外周血淋巴细胞中的表达规律,以发现外周血淋巴细胞中是否存在分子生物时钟。方法以不同光制(全黑暗和光照-黑暗交替光制)条件下饲养的 SD大鼠为研究对象,收集不同时点的外周血淋巴细胞,采用半定量RT-PCR法检测钟基因Clock及相关基因mt1 和mt2在转录本水平上的表达丰度,并用余弦软件分析各基因表达的昼夜节律特征。结果发现在光照-黑暗交替光制(LD12:12)下,Clock, mt1和mt2在大鼠外周血淋巴细胞中都具有昼夜节律性表达,其峰值位相分別 为ZT21、ZT8和ZT6;在全黑暗光制(DD)下,这种节律性表达现象仍然存在,即它们的表达节律是内源性的, 其峰值位相分别位于CT7、CT9、CT11。结论外周血淋巴细胞中可能存在受褪黑素调节的分子生物钟。

【Abstract】 Objective To explore the expression patterns of Clock and clock-related genes in the peripheral lymphocytes,and to infer if a molecular circadian clock might be existed.Methods The circadian expression patterns of the Clock and melatonin receptor genes (mt1,mt2) in the peripheral lymphocytes were determined by the method of semi-quantitative RT-PCR,and then analyzed by cosine software. Results Circadian expressions of the Clock and melatonin receptor genes (mt1,mt2) were found in the light-dark regime of LD(12:12), with the peak phase at ZT21,ZT8 and ZT6,respectively;the circadian expressions were observed in the constant dark regime(DD) too, with the peak phase at CT7, CT9 and CT11,respectively.That is,the circadian expression of these genes is endogenetic.Conclusion A molecular circadian clock regulated by melatonin might be existed in the peripheral lymphocytes of rats.
